Analysis of monoclonal antibodies reactive with the porcine CD4 antigen

open access: yesVeterinary Immunology and Immunopathology, 1994
As result of the First International Swine CD Workshop, four monoclonal antibodies (mAb) (#002, 054, 118, and 127) clustered closely to the internal standard anti-porcine CD4 mAb, 74-12-4. To further characterize the relationship between these mAb, they were used in flow cytometry to inhibit binding of 74-12-4 to porcine lymphocytes. mAb #002 (74-12-4)

Tailoring Vascular‐Immune Homeostasis via Manganese‐DNA Complex‐Armed Immunogenic Extracellular Vesicles for Pancreatic Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
This study demonstrates that Mn2⁺–tumor DNA complexes encapsulated in dendritic cell (DC)– derived immunogenic extracellular vesicles (EVDC@Mn‐DNA) act as a DC‐specific cGAS– STING activator. EVDC@Mn‐DNA treatment enhances intratumoral DC activation, improves tumor vascular function, promotes CD8⁺ T cell activity, and suppresses pancreatic tumor growth,

PD‐1 Inhibits CD4+ TRM‐Mediated cDC1 Mobilization via Suppressing JAML in Human NSCLC

open access: yesAdvanced Science, EarlyView.
CD4+ tissue‐resident memory T cells (TRMs) in non‐small cell lung cancer recruit conventional type 1 dendritic cells via XCL1‐XCR1 signaling, orchestrating antitumor immunity. The costimulatory molecule JAML is essential for this process. PD‐1 blockade restores JAML expression and cDC1 mobilization, while JAML agonists synergize with anti‐PD‐1 therapy,

Regulating Tumor Metabolic Reprogramming with Biomimetic Co‐Delivery of Simvastatin and Kynureninase for Immunotherapy

open access: yesAdvanced Science, EarlyView.
After the intravenous injection of biomimetic and pH/ROS‐responsive PTSK@CRM, the nanoparticles can be accumulated in tumors and release Sim and KYNase to inhibit the tumor growth, regulate the metabolism of cholesterol and Kyn, and reverse the immunosuppressive tumor microenvironment.

Anti‐PD‐1 Nanobody‐Armored MSLN CAR‐T Therapy for Malignant Mesothelioma: Preclinical and Clinical Studies

open access: yesAdvanced Science, EarlyView.
A novel therapy using engineered immune cells (NAC‐T cells) showed promise for refractory malignant mesothelioma. Based on the encouraging preclinical data, the first‐in‐human trial is initiated, demonstrating tolerable safety and promising anti‐tumor activity (ORR 63.6%, DCR 100%, including one CR).

Changes in T-Cell and Monocyte Phenotypes In Vitro by Schistosoma mansoni Antigens in Cutaneous Leishmaniasis Patients

open access: yesJournal of Parasitology Research, 2012
High levels of proinflammatory cytokines such as IFN-γ and TNF are associated with tissue lesions in cutaneous leishmaniasis (CL). We previously demonstrated that Schistosoma mansoni antigens downmodulate the in vitro cytokine response in CL.

A Patient‐Derived Organ‐on‐Chip Platform for Modeling the Tumor Microenvironment and Drug Responses in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
Researchers have developed a patient‐derived organ‐on‐a‐chip model for pancreatic cancer by integrating cancer cells with supportive stromal and immune cells inside a microfluidic device. This system mimics the tumor microenvironment, enabling personalized testing of chemotherapy and immunotherapy, and offering new insights into how targeting ...

Single‐Cell RNA Sequencing of Human Lung Tissues Reveals Metallothionein‐Positive T Cells as a Novel Potential Marker of Susceptibility to Chronic Obstructive Pulmonary Disease

open access: yesAdvanced Science, EarlyView.
Using single‐cell RNA sequencing of lung samples and flow cytometry of peripheral blood, we identified a novel T cell subset with high metallothionein (MT) expression. These MT‐high T cells suppress CD8+ T cell cytotoxicity and are progressively depleted from healthy controls to chronic obstructive pulmonary disease (COPD) patients, positioning them as

Identification of CD4+T-Cell-Stimulating Antigens by Expression Cloning

open access: yesMethods, 1996
The antigens recognized by CD4(+) T cells are key to rational vaccine design, but have been difficult to identify due to limitations of conventional methods. A novel strategy for identifying CD4(+) T-cell-stimulating antigen genes is described here. Using antigen-specific, lacZ-expressing T cells as single-cell probes, a DNA library was screened as ...

Western Diet Inhibits FUT7‐Mediated Treg Intestinal Homing to Disrupt the Homeostasis of Intestinal Epithelial Cells in Crohn's Disease

open access: yesAdvanced Science, EarlyView.
In this study, we constructed a nanoparticle that was CD4 antibody‐mediated targeted, Fut7‐expressing plasmid‐loaded cationic liposome, namely CD4‐LDP‐Fut7. Upregulation of Fut7 expression in Tregs by using CD4‐LDP‐Fut7 can increase Treg homing to the intestine, thereby facilitating repair of the intestinal epithelial barrier and inhibiting ...
