pH‐mediated activation of the lysosomal arginine sensor SLC38A9

open access: yesFEBS Letters, EarlyView.
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...

COMPARISON BETWEEN TWO-LAYER DENSITY GRADIENT AND THREE-LAYER DENSITY GRADIENT TECHNIQUE FOR SPERM PREPARATION AT ASTER FERTILITY CLINIC, DR. HASAN SADIKIN GENERAL HOSPITAL

open access: yesInternational Journal of Integrated Health Sciences, 2014
Objective: To compare the efficacy of sperm preparation techniques modified by two layer density gradient technique (80%, 40%). Methods: Sperm preparation was performed by two- and three-layer density gradient technique by centrifugation at 500 rpm ...

Density gradient centrifugation in anglehead rotors, Anal

open access: yes, 2020
Preparative isopycnic density-gradient centrifugation is ordinarily done in high-speed swinging-bucket rotors such as the Spinco SW 39 and SW 25. In addition to requiring this special equipment, the technique is severely limited by the small number of ...

Rab14 regulates the transport of human papillomavirus to the trans‐Golgi network for infectious cell entry

open access: yesFEBS Letters, EarlyView.
This study reveals that the small GTPase Rab14 is necessary for human papillomavirus (HPV) infection and plays an essential role in the transport of virions to the trans‐Golgi network (TGN). HPV in the early endosome (EE), which harbors GTP‐bound Rab14, is transported to the TGN through the switch of Rab14 from its GTP‐bound to GDP‐bound form.

Density Gradient Centrifugation for Enrichment and Identification of GFP-tagged Chitosomal Microvesicles of Filamentous Fungi

open access: yesBio-Protocol, 2015
Density gradient centrifugation has been utilized to characterize the subcellular distribution of physiologically relevant enzymes in yeasts and filamentous fungi (Leal-Morales et al., 1988; Martínez et al., 1989; Kamada et al., 1991).

An unexpected alternative viologen electron mediator site in tungsten‐containing formate dehydrogenase

open access: yesFEBS Letters, EarlyView.
An unexpected alternative interaction site for ethyl viologen was identified in formate dehydrogenase 1 from Methylorubrum extorquens. Combined mutagenesis, kinetic analysis, and docking revealed that aromatic residues near an iron–sulfur cluster enable flavin mononucleotide‐independent electron transfer, offering a framework for engineering improved ...

Electron transfer between complexes III and IV in S. cerevisiae mitochondrial membranes

open access: yesFEBS Letters, EarlyView.
Mitochondrial oxidative phosphorylation in S. cerevisiae mitoplasts is limited by complex IV catalytic capacity, rather than two‐dimensional cytochrome c diffusion. At physiological cytochrome c : supercomplex ratios at salinity equivalent to that of 20 mm monovalent salt, activity is maximized, indicating that this low ionic strength accurately mimics

Isolation and Culture of Murine Hepatic Stellate Cells

open access: yesBio-Protocol, 2019
Hepatic stellate cells (HSCs), alternatively known as liver pericytes, can differentiate into myofibroblasts and secrete extra-cellular matrix components, thereby promoting wound healing and fibrosis.

Comparison of density gradient and single layer centrifugation of stallion spermatozoa: Yield, motility and survival

open access: yes, 2009
Reasons for performing study: A new, simpler, technique of colloidal centrifugation has recently been developed, designated single layer centrifugation (SLC).
