Expanding Chemical Space of Nucleic Acid Nanoparticles for Tunable Antiviral‐Like Immunomodulatory Responses and Potent Adjuvant Activity

open access: yesAdvanced Functional Materials, EarlyView.
We introduce a nucleic acid nanoparticle (NANP) platform designed to be rrecognized by the human innate immune system in a regulated manner. By changing chemical composition while maintaining constant architectural parameters, we identify key determinants of immunorecognition enabling the rational design of NANPs with tunable immune activation profiles

The proto-oncogene SRC phosphorylates cGAS to inhibit an antitumor immune response

open access: yesJCI Insight, 2023
Cyclic GMP-AMP synthase (cGAS) is a DNA sensor and responsible for inducing an antitumor immune response. Recent studies reveal that cGAS is frequently inhibited in cancer, and therapeutic targets to promote antitumor cGAS function remain elusive. SRC is

Variations in caffeine and chlorogenic acid contents of coffees: what are we drinking? [PDF]

open access: yes, 2014
The effect of roasting of coffee beans and the extraction of ground coffee with different volumes of hot pressurised water on the caffeine and the total caffeoylquinic acids (CQAs) content of the resultant beverages was investigated.

Bimetallic Nanoreactor Activates cGAS‐STING Pathway via mtDNA Release for Cancer Metalloimmunotherapy

open access: yesAdvanced Functional Materials, EarlyView.
A bimetallic Mn–Ca nanoreactor (MCC) is developed as a non‐nucleotide STING nanoagonist for cancer metalloimmunotherapy. MCC induces Ca2+ overload and hydroxyl radical generation, resulting in mitochondrial damage and mtDNA release. The released mtDNA cooperates with Mn2+ to robustly activate cGAS–STING signaling.

Tight nuclear tethering of cGAS is essential for preventing autoreactivity

open access: yeseLife, 2019
cGAS is an intracellular innate immune sensor that detects double-stranded DNA. The presence of billions of base pairs of genomic DNA in all nucleated cells raises the question of how cGAS is not constitutively activated.

Topoisomerase II inhibitors induce DNA damage-dependent interferon responses circumventing Ebola virus immune evasion [PDF]

open access: yes, 2017
Ebola virus (EBOV) protein VP35 inhibits production of interferon alpha/beta (IFN) by blocking RIG-I-like receptor signaling pathways, thereby promoting virus replication and pathogenesis.

A Single‐Metal‐Doped Nanoplatform for Ferroptosis‐Driven cGAS‐STING Pathway Activation in Hepatocellular Carcinoma Immunotherapy

open access: yesAdvanced Functional Materials, EarlyView.
The cGAS‐STING pathway boosts HCC antitumor immunity but lacks specific activation. Nanoplatform ZMRPF induces HCC ferroptosis via lipid ROS, releasing mtDNA. It synergizes with ZMRPF‐released Mn2⁺ to activate cGAS‐STING, amplifies antigen‐presenting cell activity, reverses HCC immunosuppression, and enables robust systemic antitumor immunity ...

Two-phase distribution in the vertical flow line of a domestic wet central heating system [PDF]

open access: yes, 2013
The theoretical and experimental aspects of bubble distribution in bubbly two-phase flow are reviewed in the context of the micro bubbles present in a domestic gas fired wet central heating system.

ZYG11B potentiates the antiviral innate immune response by enhancing cGAS-DNA binding and condensation

open access: yesCell Reports, 2023
Summary: As a key dsDNA recognition receptor, cyclic guanosine monophosphate (GMP)-AMP synthase (cGAS) plays a vital role in innate immune responses. Activated cGAS, by sensing DNA, catalyzes the synthesis of the secondary messenger cyclic GMP-AMP (cGAMP)
