Mutant KRAS‐m6A Epitranscriptome Axis Promotes Colorectal Cancer and is a Therapeutic Target

open access: yesAdvanced Science, EarlyView.
Mutant KRAS‐m6A axis promotes colorectal cancer (CRC) progression. Mutant KRAS stabilizes METTL3, leading to increased m6A‐modified BCL9L mRNA and translation of BCL9L protein. BCL9L in turn mediates TGF‐β secretion to induce differentiation of Treg and an immunosuppressive microenvironment.

Chloroquine Transport via the Malaria Parasite’s Chloroquine Resistance Transporter

open access: yesScience, 2009
Malaria Chloroquine Resistance Transporter Malaria is one of the most deadly infectious diseases in the world today, and the emergence and spread of chloroquine-resistant parasites has been a disaster for world health.

Lyssavirus Nucleoprotein Induces GPAT4‐Mediated CYB5R1 Depalmitoylation to Suppress Ferroptosis and Promote Viral Replication

open access: yesAdvanced Science, EarlyView.
Lipid metabolism plays a vital role in viral infections. Here, we reveal that the lyssavirus N protein regulates GPAT4 to induce lipid droplet biogenesis, restricting CYB5R1 palmitoylation and reducing ferroptosis susceptibility to ultimately promote viral replication.

Targeted Lysosomal Degradation of Extracellular and Membrane Proteins: From Receptor Hijacking to Programmable Endolysosomal Routing

open access: yesAdvanced Science, EarlyView.
Targeted chimeric degraders engage extracellular or membrane proteins and endocytosable cell‐surface receptors, forming ternary complexes that induce receptor‐mediated internalization. Following endocytosis, post‐endocytic sorting—not internalization alone—determines target fate.

Beyond Biomimetics: Pathology‐Informed Engineering Rescues Reconstituted HDL From Inflammatory Dysfunction for Sepsis Immunotherapy

open access: yesAdvanced Science, EarlyView.
A pathology‐informed engineering strategy rescues reconstituted HDL from inflammatory dysfunction in sepsis. By integrating a ceria nanozyme‐celastrol coordination complex, the platform preserves HDL structure and endotoxin handling, sustains hierarchical intracellular delivery, and enhances immunoregulation across pathological models, translating ...

Granzyme B PET Imaging Reveals Lgmn+ Macrophage‐Mediated Immune Evasion and Guides Immunotherapy in EGFR‐TKI‐Resistant NSCLC

open access: yesAdvanced Science, EarlyView.
Graphical illustration of 68Ga‐GZB PET/CT imaging reveals immune dynamics post‐EGFR‐TKI resistance in NSCLC; Lgmn+ macrophages‐derived Lgmn‐containing extracellular vesicles inhibit glycolysis, induce CD8+ T cell exhaustion, and targeting Lgmn restores immunotherapy sensitivity.

Use of chloroquine in uncomplicated falciparum malaria chemotherapy : The past, the present and the future

open access: yesSahel Medical Journal, 2010
Chloroquine is a 4-aminoquinoline discovered over five decades ago for treatment of uncomplicated malaria. It was widely used as first line treatment and prophylaxis for individuals going into malaria endemic regions.

Transcripts of the multidrug resistance genes in chloroquine-sensitive and chloroquine-resistantPlasmodium falciparum

open access: yesParasitology, 1993
SUMMARYHomologues of the mammalian multidrug resistance gene have been identified in isolates and clones ofPlasmodium falciparumand designatedpfmdr1andpfmdr2.Mutations inpfmdr1have been associated with chloroquine resistance but confirmation could not be obtained in a genetic cross.

The rationale and plan for creating a World Antimalarial Resistance Network (WARN) [PDF]

open access: yes, 2007
Drug resistant malaria was a major factor contributing to the failure of a worldwide campaign to eradicate malaria in the last century, and now threatens the large investment being made by the global community in the rollout of effective new drug ...

Malaria Control Strategies in the Kilombero Valley, Tanzania [PDF]

open access: yes, 2000
Malaria is major public health problem in Tanzania and increasing trends have been observed in the last two decades. A significant consequence of repeated malaria infections in high transmission areas is anaemia in very young children.
