Preparative synthesis of prephenate from chorismate by chorismate mutase immobilized on decylamine agarose

Abstract Chorismate mutase (EC 5.4.99.5) from Streptomyces aureofaciens is bound virtually irreversibly by n -decylamine-substituted agarose and remains active when immobilized by hydrophobic binding. The enzyme is not eluted by buffers containing 1 m NaCl or 50% ethylene glycol.

Chorismate synthase of Neurospora crassa: A flavoprotein

Abstract Chorismate synthase is purified from Neurospora crassa. The final step is accomplished by preparative electrophoresis. Its purity is estimated at ≥90% on the basis of analytical polyacrylamide gel electrophoresis. The enzyme appears to be active in at least two multimeric states, with a subunit molecular weight of ~55,000.

1. Chorismate mutase, the first enzyme of the terminal biosynthetic pathway of phenylalanine and tyrosine has been purified 10 fold fromStreptomyces venezuelae. 2. The catalytic activity has a broad optimum between pH 6.6 and 7. The initial velocity data follow regular Michaelis-Menten kinetics with aKm of 3.8×10-4 M.

The Saccharomyces cerevisiae ARO7 gene was cloned by screening a wild-type gene bank for complementation of an aro7 auxotrophic mutant. In vitro mutagenesis of the isolated plasmid (pJFB1) gave several transformants resistant to levels of the phenylalanine analogue 2-thienylalanine inhibitory to the wild-type transformant.
