Re-defining the chromatin loop domain

Cytogenetic and Genome Research, 2001
It is commonly accepted that the loop domain represents the basic structural unit of eukaryotic chromatin associated with DNA replication, gene expression and higher order packaging. However, molecular-cytological information defining the loop domain is lacking. There are gaps in our knowledge of the loop structure and how it regulates gene expression.

The role of chromatin loop extrusion in antibody diversification

Nature Reviews Immunology, 2022
Cohesin mediates chromatin loop formation across the genome by extruding chromatin between convergently oriented CTCF-binding elements. Recent studies indicate that cohesin-mediated loop extrusion in developing B cells presents immunoglobulin heavy chain (Igh) variable (V), diversity (D) and joining (J) gene segments to RAG endonuclease through a ...

Chromatin loops, illegitimate recombination, and genome evolution

BioEssays, 2009
AbstractChromosomal rearrangements frequently occur at specific places (“hot spots”) in the genome. These recombination hot spots are usually separated by 50–100 kb regions of DNA that are rarely involved in rearrangements. It is quite likely that there is a correlation between the above‐mentioned distances and the average size of DNA loops fixed at ...

Analysis of Chromatin Interaction and Accessibility by Trac-Looping

2023
Spatial organization of the genome modulates pivotal biological processes. The emerging new technologies have provided novel insights into genome structure and its role in regulating cell activities. To examine the genome-wide chromatin interactions at accessible chromatin regions, we developed a DNA transposase-mediated analysis of chromatin looping ...

BIOLOGICAL CONSEQUENCES OF CHROMATIN LOOPING IN PERICENTRIC CHROMATIN

2018
During mitosis, replicated sister chromatids are attached to opposite sides of a microtubule spindle at their centromeres in a process called biorientation. The proteinaceous structure that links centromeres, a region of the chromosome, to the spindle is called the kinetochore.

Chromatin loops and transcriptional regulation.

Critical reviews in eukaryotic gene expression, 1992
The existence of a supermolecular structure in the eukaryotic nucleus, involving loops of chromatin attached at irregularly spaced points to the nuclear matrix, is now well established. Quite a bit is known concerning the DNA sequences involved in the attachment. However, the function of this highly organized structure remains largely unknown.
