Ciguatoxin and Ciguatera

2015
Ciguatera is a disease caused by the consumption of fishes from tropical and subtropical waters that have accumulated lipophilic sodium channel activator toxins known as ciguatoxins (CTXs) to levels sufficient to cause human poisoning. Consumption of these temperature-stable, orally active polycyclic ether compounds leads to the activation of neuronal ...

Optimization of ciguatoxin extraction method from blood for Pacific ciguatoxin (P-CTX-1)

Toxicon, 2007
Ciguatera diagnosis relies on clinical observations associated with a recent consumption of fish. Although needed, direct confirmation of exposure in subjects showing ciguatera disease symptoms is currently unavailable. We previously reported that ciguatoxins were measurable in the blood of mice exposed to extracts of Pacific ciguatoxins isolated from ...

Synthesis and Structural Implication of the JKLMN-Ring Fragment of Caribbean Ciguatoxin C-CTX-1.

Journal of Organic Chemistry, 2021
Synthesis of the JKLMN-ring fragment of Caribbean ciguatoxin C-CTX-1, the causative toxin of ciguatera fish poisoning in the Caribbean Sea and the Northeast Atlantic areas, is described in detail.

Recovery of ciguatoxin from fish flesh

Toxicon, 1993
A mouse bioassay, validated for the quantification of ciguatoxin in up to 20 mg of ether extract from fish flesh, revealed that 63 +/- 14% of spiked ciguatoxin was recovered using a standard extraction procedure. Except for extracts from the least toxic of ciguateric fish (0.1-0.5 nmol ciguatoxin-1/kg fish), signs in mice of intoxication by ciguatoxin (

Synthesis of the MN Ring of Caribbean Ciguatoxin C-CTX-1 via Desymmetrization by Acetal Formation.

Organic Letters
The MN ring of Caribbean ciguatoxin C-CTX-1 was synthesized from a meso-syn-2,7-dimethyloxepane derivative corresponding to the M ring via desymmetrization by acetal formation with a camphor derivative, followed by construction of the N ring via the ...

Analytical challenges to ciguatoxins

Current Opinion in Food Science, 2017
Ciguatera fish poisoning (CFP) is a form of food poisoning caused by consumption of varieties of toxic ciguatera fish species from tropical and subtropical waters. In the present review, significant features and perspective of LC/MS methods for CTXs as confirmatory methods are reviewed on the discussion for one of the best LC/MS/MS method reported by ...

Total Synthesis of Ciguatoxin and 51-HydroxyCTX3C

Journal of the American Chemical Society, 2006
Ciguatoxins, the principal causative toxins of ciguatera seafood poisoning, are large ladder-like polycyclic ethers with the 13 ether rings ranging from five- to nine-membered. In this paper, we describe the total synthesis of the two most toxic members of the ciguatoxin family, ciguatoxin 1 and 51-hydroxyCTX3C 2, based on a unified synthetic strategy.

A radioimmunoassay for the detection of ciguatoxin

Toxicon, 1977
Abstract A practical, sensitive, simple and relatively specific radioimmunoassay test for the assay of ciguatoxin directly from natural sources has been developed. The radioimmunoassay test has been utilized to distinguish the toxic fishes in human ciguatera poisoning outbreaks from the nontoxic fishes.

Ciguatoxin: developing the methodology for total synthesis

Natural Product Reports, 2010
Covering: up to 2009 The ciguatoxins are sodium channel toxins from the marine dinoflagellate Gambierdiscus toxicus, isolated in 1984. In this review, we summarize the principal concepts and recently developed methodologies for the synthesis of the ciguatoxins, including C–C bond formation by silylacetylene and cobalt-mediated cyclization.
