Structure Activity Relationship Studies around DB18, a Potent and Selective Inhibitor of CLK Kinases

open access: yesMolecules, 2022
Three series of our lead CLK1 inhibitor DB18 have been designed, synthetized and tested against CLKs and DYRK1A kinases. Their cytotoxicity was subsequently measured on seven representative cancer cell lines.

The Identification of Prognostic and Metastatic Alternative Splicing in Skin Cutaneous Melanoma

open access: yesCancer Control, 2022
Skin cutaneous melanoma (SKCM) is a type of highly invasive cancer originated from melanocytes. It is reported that aberrant alternative splicing (AS) plays an important role in the neoplasia and metastasis of many types of cancer.

CLK1 reorganizes the splicing factor U1-70K for early spliceosomal protein assembly [PDF]

open access: yesProceedings of the National Academy of Sciences, 2021
Significance Proteome size is greatly enhanced through alternative mRNA splicing, a process catalyzed by the macromolecular spliceosome. The initial step in spliceosome assembly involves the binding of U1 snRNP to the 5′ splice site in mRNA. SR proteins ensure the fidelity of this step by simultaneously binding to mRNA and U1-70K, a protein ...

Design and biological evaluation of substituted 5,7-dihydro-6H-indolo[2,3-c]quinolin-6-one as novel selective Haspin inhibitors

open access: yesJournal of Enzyme Inhibition and Medicinal Chemistry, 2022
A library of substituted indolo[2,3-c]quinolone-6-ones was developed as simplified Lamellarin isosters. Synthesis was achieved from indole after a four-step pathway sequence involving iodination, a Suzuki-Miyaura cross-coupling reaction, and a reduction ...

Controlling clots with Clk1 [PDF]

open access: yesThe Journal of Experimental Medicine, 2006
Platelets are critical blood cells involved in clotting. An early step in clot construction requires tissue factor (TF), but whether platelets make TF was unknown. New work by Schwertz et al. (page 2433) reveals that these nuclei-free cells doproduce TF by cytoplasmic splicing and further identify the kinase Clk1 as a splicing activator.

A CLK1-KKT2 Signaling Pathway Regulating Kinetochore Assembly in Trypanosoma brucei

open access: yesmBio, 2021
In eukaryotic cells, kinetochores are large protein complexes that link chromosomes to dynamic microtubule tips, ensuring proper segregation and genomic stability during cell division.

An extensive program of periodic alternative splicing linked to cell cycle progression

open access: yeseLife, 2016
Progression through the mitotic cell cycle requires periodic regulation of gene function at the levels of transcription, translation, protein-protein interactions, post-translational modification and degradation. However, the role of alternative splicing

Lectin complement protein Collectin 11 (CL-K1) and susceptibility to urinary schistosomiasis. [PDF]

open access: yesPLoS Neglected Tropical Diseases, 2015
BACKGROUND:Urinary Schistosomiasis is a neglected tropical disease endemic in many sub Saharan -African countries. Collectin Kidney 1 (CL-K1, encoded by COLEC11 on chromosome 2p25.3), a member of the vertebrate C-type lectin super family, has recently ...

Data_Sheet_1_Inhibition of a Novel CLK1-THRAP3-PPARγ Axis Improves Insulin Sensitivity.xlsx

open access: yes, 2021
Increasing energy expenditure by promoting “browning” in adipose tissues is a promising strategy to prevent obesity and associated diabetes. To uncover potential targets of cold exposure, which induces energy expenditure, we performed phosphoproteomics ...

MAPKs and acetyl-CoA are associated with Curvularia lunata pathogenicity and toxin production in maize

open access: yesJournal of Integrative Agriculture, 2018
Mitogen-activated protein kinase (MAPK) cascades play an important role in extracellular signal transduction and are involved in the pathogenicity of fungal pathogens to host plants.
