Insights into Co-Amorphous Systems in Therapeutic Drug Delivery
Therapeutic Delivery, 2021Co-amorphous (CAM) systems are promising drug-delivery systems in the arena of therapeutic drug delivery, addressing the poor aqueous solubility of drugs by enhancing solubility and thereby improving the oral bioavailability and therapeutic effect of the drug.

Dipeptides as co-formers in co-amorphous systems
European Journal of Pharmaceutics and Biopharmaceutics, 2019Drug-amino acid co-amorphous systems have become increasingly well-investigated systems to improve dissolution rate of poorly water-soluble drugs. In this study, dipeptides were investigated as co-formers for co-amorphous systems based on the hypothesis that dipeptides might combine the inherent properties of the two included amino acids.

Aspartame as a co-former in co-amorphous systems
International Journal of Pharmaceutics, 2018Co-amorphous drug delivery systems are a promising approach to improve the dissolution rate and therefore potentially the oral bioavailability of poorly-water soluble drugs. Several low molecular weight excipients, for instance amino acids, have previously been shown to stabilize the amorphous form and increase the dissolution rate of drugs.

Platensimycin-berberine chloride co-amorphous drug system: Sustained release and prolonged half-life
European Journal of Pharmaceutics and Biopharmaceutics, 2022Co-amorphous technology is an emerging approach for pharmaceutical engineering of drugs and drug leads with improved physicochemical properties and bioavailability. Platensimycin (PTM) is a promising natural antibiotic lead that acts on bacterial fatty acid synthase and exhibits excellent antibacterial activity.

Spray drying is an increasingly used particle engineering technique for the production of dry powders for inhalation. However, the amorphous nature of most spray-dried particles remains a big challenge affecting both the chemical and the physical stability of the dried particles.

There is a great interest to develop co-amorphous drug delivery systems to enhance the solubility of biopharmaceutics classification system (BCS) class II and IV drugs. However, most reported systems only resulted in severalfold solubility improvement. Tranilast (TRA) is an anti-allergic drug used to treat bronchial asthma and allergic rhinitis.

Co-amorphization is a promising approach to stabilize drugs in the amorphous form. Olanzapine, a poorly water-soluble drug was used in this study. Sulfonic acids (saccharin, cyclamic acid and acesulfame), free and in salt forms, were used as co-formers and compared with carboxylic acids commonly used in the preparation of co-amorphous systems.

[The development of co-amorphous drug systems].
Yao xue xue bao = Acta pharmaceutica Sinica, 2016Converting two poorly water-soluble crystalline drugs to co-amorphous drug systems by ball milling, quench-cooling, or cryo-milling method can improve stability of the drug, enhance dissolution rates, and reduce adverse reactions of the single drug. Co-amorphous system has been used to solve problems of co-administration of medicines.

Influence of variation in molar ratio on co-amorphous drug-amino acid systems
European Journal of Pharmaceutics and Biopharmaceutics, 2016Molecular interactions were investigated within four different co-amorphous drug-amino acid systems, namely indomethacin-tryptophan (Ind-Trp), furosemide-tryptophan (Fur-Trp), indomethacin-arginine (Ind-Arg) and furosemide-arginine (Fur-Arg). The co-amorphous systems were prepared by ball milling for 90min at different molar ratios and analyzed by XRPD
