Cobamide-producing microbes as a model for understanding general nutritional interdependencies in soil food webs [PDF]

open access: yesNature Communications
Nutrient crossfeeding critically governs microbiome–host interactions and ecosystem stability. Cobamides, synthesized only by prokaryotes, offer a powerful and tractable model for studying nutrient-mediated interdependencies in soil food webs; however ...

Structural studies of the phosphoribosyltransferase involved in cobamide biosynthesis in methanogenic archaea and cyanobacteria [PDF]

open access: yesScientific Reports, 2022
Cobamides (Cbas) are coenzymes used by cells across all domains of life, but de novo synthesis is only found in some bacteria and archaea. Five enzymes assemble the nucleotide loop in the alpha phase of the corrin ring. Condensation of the activated ring

Reduction of Cobamides by Reduced Ferredoxin

open access: yesJournal of Biological Chemistry, 1966
Reduced ferredoxin is capable of reducing nonenzymatically cyano-B12. Spectral studies have shown that B12r (a 1-electron reduced derivative of the vitamin) accumulates in the reaction although the formation of methyl-B12 was observed upon the addition of methyl iodide to the incubation.

Direct Cobamide Remodeling via Additional Function of Cobamide Biosynthesis Protein CobS from Vibrio cholerae. [PDF]

open access: yesJ Bacteriol, 2021
Cobamides are widespread cofactors that mediate metabolic interactions in complex microbial communities. Few studies directly examine cobamide profiles, but several have shown that mammalian gastrointestinal tracts are rich in cobamide analogs.

Corrinoid salvaging and cobamide remodeling in bacteria and archaea. [PDF]

open access: yesJ Bacteriol
ABSTRACT Cobamides (Cbas) are cobalt-containing cyclic tetrapyrroles used by cells from all domains of life as co-catalyst of diverse reactions. There are several structural features that distinguish Cbas from one another.

Naturally occurring cobalamin (B12) analogs can function as cofactors for human methylmalonyl-CoA mutase. [PDF]

open access: yesBiochimie, 2021
Cobalamin, commonly known as vitamin B12, is an essential micronutrient for humans because of its role as an enzyme cofactor. Cobalamin is one of over a dozen structurally related compounds - cobamides - that are found in certain foods and are produced ...

Evidence for corrin biosynthesis in the last universal common ancestor. [PDF]

open access: yesFEBS J
Cobalamin biosynthesis can be traced back to the last universal common ancestor. After a stage of chemical evolution of the acetyl‐CoA pathway, the enzymatic synthesis of cobalamin evolved, allowing life to be independent of the insoluble solid‐state catalysts that tethered primordial CO2 assimilation to the Earth's crust, suggesting a role for corrin ...

Evolutionarily divergent DUF4465 domains have a common vitamin B12-binding function. [PDF]

open access: yesFEBS Open Bio
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

Elevated Levels of an Enzyme Involved in Coenzyme B12 Biosynthesis Kills Escherichia coli

open access: yesmBio, 2022
Cobamides are cobalt-containing cyclic tetrapyrroles involved in the metabolism of organisms from all domains of life but produced de novo only by some bacteria and archaea.

Characterization of Vitamin B12 Compounds in Fermented Poultry Manure Fertilizers

open access: yesAgriculture, 2021
(1) Background: Currently, no data are available on the vitamin B12 content of an organic fertilizer product, viz. fermented poultry manure, or whether the organic fertilizer product contains vitamin B12 or inactive corrinoids (or both).
