Pharmaceutical Cocrystals and Their Physicochemical Properties [PDF]

open access: yesCrystal Growth & Design, 2009
This review article will highlight and discuss the advances made over the last 10 years pertaining to physical and chemical property improvements through pharmaceutical cocrystals and, hopefully, draw closer the fields of crystal engineering and pharmaceutical sciences.

Chiral (Stereoselective) Drugs, Asymmetric Synthesis, and Racemic Resolution Methods

open access: yesChirality, Volume 38, Issue 7, July 2026.
Chirality is crucial in drug development since both biological targets in the organism and the majority of pharmaceutical compounds are chiral. The synthesis and resolution of chiral compounds are critical steps while developing chiral drugs. Asymmetric synthesis and racemic resolution are the two most common methods for obtaining enantiopure drugs ...

Phosphoryl Halogen and Hydrogen Bonding Controls the Solid‐State Structures of Aryl Halide‐Containing Phosphonates

open access: yesEuropean Journal of Inorganic Chemistry, Volume 29, Issue 17, 17 June 2026.
Intramolecular (RO)2PO→Halogen interaction versus intermolecular (RO)2PO···HOH hydrogen bonds. Although the O···I distance is shorter than the sum of the van der Waals radii of the atoms involved, there is no covalency involved in this interaction.

Direct and Telescopic Mechanochemical Synthesis of Higher-order Organic-Inorganic Hybrid Cocrystals: Tuning Order, Functionality and Size in Cocrystal Design

open access: yes, 2021
The ability to rationally design and predictably construct crystalline solids has been the hallmark of crystal engineering research over the past two decades. When building higher-order multicomponent cocrystals (i.e.

Pharmaceutical Cocrystals of Niclosamide

open access: yes, 2016
Niclosamide (NCL) is an anthelmintic BCS class II drug of low solubility and high permeability. Pharmaceutical cocrystals of NCL were prepared with GRAS molecules, such as caffeine (CAF), urea (URE), p-aminobenzoic acid (PABA), theophylline (THPH ...

Mechanochemical Preparations of Anion Coordinated Architectures Based on 3‐Iodoethynylpyridine and 3‐Iodoethynylbenzoic Acid

open access: yesChemistryOpen, 2019
The halogen bond has previously been explored as a versatile tool in crystal engineering and anion coordination chemistry, with mechanochemical synthetic techniques having been shown to provide convenient routes towards cocrystals. In an effort to expand

Structure‐Based Design, Synthesis, and Evaluation of Novel Ponatinib Derivatives With a Significantly Altered Selectivity Profile

open access: yesChemMedChem, Volume 21, Issue 11, 15 June 2026.
We developed new derivatives of the cancer drug ponatinib, significantly reducing and altering its spectrum of target kinases. Compound 5 retained the efficacy in inhibiting the colony formation of MDA‐MB‐231 cells and acted primarily on B‐Raf and Flt‐1 as its main targets in a kinase panel.

Dissolution Advantage of Nitazoxanide Cocrystals in the Presence of Cellulosic Polymers

open access: yesPharmaceutics, 2019
The effect of hydroxypropyl methylcellulose (HPMC) and methylcellulose (Methocel® 60 HG) on the dissolution behavior of two cocrystals derived from nitazoxanide (NTZ), viz., nitazoxanide-glutaric acid (NTZ-GLU, 1:1) and nitazoxanide-succinic acid ...

π‐Extended Chiral Molecular Triangular Prisms Exhibiting Boosted Circularly Polarized Luminescence Upon Self‐Assembly

open access: yesAggregate, Volume 7, Issue 6, June 2026.
A pair of π‐extended chiral triangular prisms based on pyromellitic diimide is synthesized from enantiopure macrocyclic precursors. They exhibit orange fluorescence and solvent‐dependent self‐assembly, enabling chiroptical inversion and amplification with |glum| values up to 1.15 × 10−2. Single‐crystal structures reveal solvent‐controlled conformations
