pH‐mediated activation of the lysosomal arginine sensor SLC38A9

open access: yesFEBS Letters, EarlyView.
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...

Optimizing Bus Driver Scheduling: A Set Covering Approach for Reducing Transportation Costs

open access: yesApplied System Innovation
Cutting operational costs is a critical component for transportation agencies. To reduce these costs, agencies must optimize their scheduling. Typically, the total operating costs of transport include vehicle expenses and driver wages. Solving such tasks

Abordagens complementares para problemas de p-medianas Complementary approaches for p-median location problems

open access: yesProduction, 2003
A localização de p-medianas é um problema clássico de otimização combinatória. O objetivo é localizar em uma rede p nós (denominados medianas), de forma a minimizar a soma das distâncias de cada nó de demanda até sua mediana mais próxima.

Degradation mechanism of the von Willebrand factor A2 domain by nattokinase

open access: yesFEBS Letters, EarlyView.
Nattokinase, a natto‐derived protease, exhibits potent antithrombotic effects. This study demonstrates that nattokinase directly cleaves the von Willebrand factor (vWF) A2 domain in vitro. Unlike the native regulator ADAMTS13, nattokinase degrades folded vWF independently of shear stress.

A Constraint Programming based column generation approach to nurse rostering problems

open access: yes, 2012
This paper presents our investigations on a hybrid constraint programming based column generation (CP–CG) approach to nurse rostering problems. We present a complete model to formulate all the complex real-world constraints in several benchmark nurse ...

An enhanced concave program relaxation for choice network revenue management [PDF]

open access: yes, 2013
The network choice revenue management problem models customers as choosing from an offer set, and the firm decides the best subset to offer at any given moment to maximize expected revenue.

Optimization Models for Reducing Off-Cuts of Raw Materials in Construction Site

open access: yesMathematics, 2022
More than ten billion tons of construction waste are generated every year in the world. The large volume of construction waste not only increases costs for contractors, but also poses a threat to the environment.

Modulation of Homer1 EVH1 domain internal dynamics by putative autism‐associated mutations

open access: yesFEBS Letters, EarlyView.
The putative autism‐associated M65I and S97L variants of the EVH1 domain of the postsynaptic scaffold protein Homer1 do not exhibit substantial changes in their overall structure or partner binding. Both of them, but especially the M65I variant, show altered internal dynamics relative to the wild‐type domain on the μs‐ms timescale, indicated by the ...

An unexpected alternative viologen electron mediator site in tungsten‐containing formate dehydrogenase

open access: yesFEBS Letters, EarlyView.
An unexpected alternative interaction site for ethyl viologen was identified in formate dehydrogenase 1 from Methylorubrum extorquens. Combined mutagenesis, kinetic analysis, and docking revealed that aromatic residues near an iron–sulfur cluster enable flavin mononucleotide‐independent electron transfer, offering a framework for engineering improved ...

Salmonella lipopolysaccharide‐containing supported lipid bilayers as platforms to study bacteriophage interactions

open access: yesFEBS Letters, EarlyView.
We present robust protocols for the preparation of supported lipid bilayers (SLBs) incorporating either Salmonella smooth LPS or outer membrane vesicles (OMVs). We use a combination of quartz crystal microbalance with dissipation (QCM‐D) and fluorescence microscopy to both characterize the SLBs of various compositions and to probe their interactions ...
