Continuous Evolution System Based on Toxin–Antitoxin System Combined with Base Deaminase Accelerates the In Vivo Modification of Target Proteins

open access: yesAdvanced Science, EarlyView.
The promoted Escherichia coli‐assisted continuous evolution (PEACE) system operates through a stringent three step genetic circuit: target gene mutation via a deaminase–T7 RNA polymerase (T7 RNAP) fusion, coupling variant function to antitoxin expression, and growth based selection of surviving cells.

Complement dependent cytotoxicity in chronic lymphocytic leukemia: ofatumumab enhances alemtuzumab complement dependent cytotoxicity and reveals cells resistant to activated complement [PDF]

open access: yesLeukemia and Lymphoma, 2012
Complement dependent cytotoxicity (CDC) is an important mechanism of action for monoclonal antibodies (mAbs) used in the treatment of chronic lymphocytic leukemia (CLL). We hypothesized that alemtuzumab (ALM) mediated CDC would be increased by the addition of ofatumumab (OFA).

Microcalorimetry as a tool for the detection of complement-dependent cytotoxicity

Journal of Immunological Methods, 1985
Microcalorimetry using a 4-channel static ampoule microcalorimeter of thermopile type has been evaluated as a tool for the detection of complement-dependent cytotoxicity against the surface antigens of living cells. Cytotoxic reactions mediated by a rabbit antiserum against human white blood cells and by 2 different monoclonal antibodies recognizing a ...

Complement-dependent phagocytosis as a regulator of antibody-dependent cellular cytotoxicity

The Journal of Immunology, 2022
Abstract While monoclonal antibodies (mAb), such as αCD20, have significantly improved patient outcomes for B cell malignancies, patient responses vary and drug resistance can occur. One proposed limitation is the reduction of innate immune effector functions such as antibody dependent cellular phagocytosis (ADCP) – the principal ...

Complement-Dependent Cytotoxicity Assay

Cold Spring Harbor Protocols, 2018
A useful feature of therapeutic antibodies is the ability to kill the cells to which they bind. Antibodies are capable of mediating cell killing in a variety of ways. Apoptosis, complement-mediated mechanisms, and antibody-dependent cellular cytotoxicity are all effects that can be assayed to characterize lead antibody candidates.

Complement-Dependent Cytotoxicity of an Antibody to Melanin in Radioimmunotherapy of Metastatic Melanoma

Immunotherapy, 2013
Novel treatments for metastatic melanoma are urgently needed. We developed radioimmunotherapy of metastatic melanoma using 6D2 monoclonal antibody (mAb) to melanin with encouraging therapeutic results, preclinically and in patients. We observed tumor suppression with the unlabeled 6D2 mAb and investigated its tumoricidal mechanisms.
