Setting up multivariate specifications on critical raw material attributes to ensure consistent drug dissolution from high drug-load sustained-release matrix tablet

open access: yesDrug Development and Industrial Pharmacy, 2018
The purpose of this study was to describe the raw material variability that influenced the in-vitro dissolution behavior of high drug-load sustained-release matrix tablet and to ensure the consistent quality of the final product. The Panax notoginseng saponins (PNS) – hydroxypropyl methylcellulose – anhydrous lactose – magnesium stearate (57:20:23:0.5%,

Spatial and single‐nuclei transcriptomics reveals idiosyncratic and generic patterns in papillary and anaplastic thyroid cancers

open access: yesMolecular Oncology, EarlyView.
Matched spatial transcriptomics and single‐nuclei RNA‐seq were generated for anaplastic and BRAFV600E papillary thyroid cancers revealing generic and tumor‐specific states occurring in cancer cells and in the tumor microenvironment. In this context, cancer dedifferentiation mirrored organoid maturation through ordered thyroid marker gain/loss ...

Stakeholder engagement to identify barriers to implementation and inform the development of point-of-care diagnostics for TB

open access: yesIJTLD Open
BACKGROUND: Rapid TB diagnosis is crucial for improving outcomes and reducing transmission, yet nearly one third of cases remain undiagnosed or unreported. Point-of-care (POC) diagnostics are critical for bridging this gap.

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Rational Function-Based Approach for Integrating Tableting Reduced-Order Models with Upstream Unit Operations: Lubricants and Glidants Case Study

open access: yesPharmaceuticals
Background/Objectives: Glidants and lubricants are commonly used pharmaceutical excipients that enhance powder flowability and reduce inter-particle friction, respectively, but they also negatively impact critical quality attributes such as tablet ...

Circulating microRNA signatures of cachexia and cancer in Canis familiaris as a comparative oncology model for human disease

open access: yesMolecular Oncology, EarlyView.
Circulating microRNAs as biomarkers of cachexia and sex‐specific cancer in senior dogs. In 25 client‐owned dogs, four circulating miRNAs (miR‐15a, miR‐15b, miR‐16, miR‐140) were downregulated in cachexia, with miR‐16 the strongest individual biomarker (AUC = 0.899).

The Effects of Trivial Attributes on Choice of Food Products [PDF]

open access: yes
Trivial or irrelevant attributes are defined as attributes that do not create a meaningful difference in a brand’s performance. The objective of this paper is to determine if and how trivial attributes affect consumers in their choice of variety/brands ...

Selective laser sintering responses of keratin-based bio-polymer composites

open access: yesMaterials & Design, 2019
Keratin-based materials have unique biological and mechanical attributes due to the protein's molecular structure and organisation. The current study evaluates the possible consolidation of keratin powders by selective laser sintering (SLS), a key ...

Epigenetic silencing of the liver‐specific lncRNA LUNAR promotes liver cancer progression via NOTCH activation

open access: yesMolecular Oncology, EarlyView.
LUNAR is a liver‐specific long noncoding RNA (lncRNA) that is highly expressed in normal liver but becomes epigenetically silenced in hepatocellular carcinoma through promoter hypermethylation. Loss of LUNAR is associated with NOTCH activation, epithelial–mesenchymal transition, and metastasis, whereas restoring LUNAR restrains metastatic progression ...

Single‐cell DNA methylation profiling: Technologies, computation, and applications in precision oncology

open access: yesMolecular Oncology, EarlyView.
Single‐cell DNA methylation (scDNAme) profiling maps epimutational clonal evolution, revealing mechanisms of malignancy and therapeutic resistance across diverse cancer types. By providing a high‐resolution landscape of intratumoral heterogeneity, these technologies empower precise patient stratification, guide the development of enhanced ...
