The Burden of Adverse Drug Reactions in Africa in the Context of Pharmacogenetics‐Based Clinical Guidelines

open access: yesClinical Pharmacology &Therapeutics, Volume 119, Issue 5, Page 1382-1390, May 2026.
Genetic variation has a significant impact on patients' response to medicines. Variations in important pharmacogenes have been evaluated in several studies and consequently led to international guidelines from clinical pharmacogenomics consortia. However, there are limited examples of these being implemented across the African continent despite the ...

Comments to the Article Wu Y, Völler S, Goulooze SC, et al. A Novel Maturation Equation for Hepatic Clearance Across Preterm, Term Neonates, Children, and Adults: Application to Paracetamol and Its Metabolite. J Clin Pharmacol. 2025;65(12):1829‐1843. doi: 10.1002/jcph.70080. PMID: 40910464; PMCID: PMC12649303

open access: yesThe Journal of Clinical Pharmacology, Volume 66, Issue 5, May 2026.
Abstract A recent article in this journal by Wu et al presented a comprehensive model for the maturation of paracetamol clearance from birth to adulthood using pooled plasma and urinary data of the parent drug and its metabolites from 298 subjects. The modeling showed that the oxidative bioactivation to hepatotoxic metabolite N‐acetyl‐p‐benzoquinone ...

Contrasting exome constancy and regulatory region variation in the gene encoding CYP3A4: an examination of the extent and potential implications. [PDF]

open access: yes, 2016
OBJECTIVE: CYP3A4 expression varies up to 100-fold among individuals, and, to date, genetic causes remain elusive. As a major drug-metabolizing enzyme, elucidation of such genetic causes would increase the potential for introducing personalized dose ...

HIV treatment outcomes in Uganda : the impact of baseline characteristics and variability in pharmacokinetics and pharmacogenetics of antiretroviral drugs [PDF]

open access: yes, 2013
Guidelines specify criteria for initiation and monitoring of antiretroviral treatment (ART), including options for first line regimens. Immunological progress remains a widely used form of HIV/ART monitoring and efavirenz, a preferred first line ...

Predicting Pharmacokinetic Variability and Drug Interaction Risk Using Omics‐Based Biomarkers

open access: yesClinical and Translational Science, Volume 19, Issue 5, May 2026.
ABSTRACT Interindividual variability in drug pharmacokinetics and susceptibility to drug–drug interactions remain major barriers in precision dosing, particularly for narrow therapeutic index drugs. While genetic factors contribute, much variability arises from dynamic influences such as physiology, disease, age, diet, microbiome, and concomitant ...

Pharmacogenetics of Mycophenolic Acid in Patients of African Descent: Underrepresentation of African Data

open access: yesClinical and Translational Science, Volume 19, Issue 5, May 2026.
ABSTRACT Mycophenolic acid (MPA) is a potent antiproliferative immunosuppressive agent used to prevent organ transplant rejection and to treat various immune‐mediated diseases. MPA is the active metabolite formed from the biotransformation of the prodrugs mycophenolate mofetil (MMF) or enteric‐coated mycophenolate sodium (EC‐MPS).

Impact of Concomitant Metamizole Treatment on the Exposure of Mould‐Active Triazoles

open access: yesMycoses, Volume 69, Issue 5, May 2026.
ABSTRACT Aim Metamizole is an analgesic drug with moderate cytochrome P450 (CYP) inductive properties. The antifungal triazoles are metabolized by several CYP enzymes, but the interaction with metamizole remains poorly described. We investigated the influence of metamizole on the exposure of voriconazole, isavuconazole, and posaconazole.

Dosing, Safety, and Tolerability of Extended‐Release Tacrolimus in Pediatric and Young Adult Solid Organ Transplant Recipients

open access: yesPediatric Transplantation, Volume 30, Issue 5, May 2026.
Extended‐release tacrolimus (LCPT) was safe and well‐tolerated among 91 children and young adult solid organ transplant patients at our hospital. However, the dosing ratio of LCPT:immediate‐release tacrolimus (mg/day) varied widely across patients, highlighting the importance of close therapeutic drug monitoring when transitioning to LCPT.
