Oligopeptides/DNA Coacervate Droplets as Macromolecular Delivery Microcarriers

open access: yesAdvanced Science, EarlyView.
Oligopeptide–DNA coacervates formed via liquid–liquid phase separation serve as programmable carriers for macromolecular cargos, entering cells through lipid raft–associated pathways and subsequently undergoing enzyme triggered intracellular disassembly driven by DNA degradation processes, thereby enabling efficient, controlled, and spatially defined ...

Single-Payload Bioconjugation to Native Antibodies And A General Method for the Self-Immolative Release of Amide-Containing Molecules [PDF]

open access: yes
Antibody-drug conjugates (ADCs) have emerged as a powerful form of targeted cancer therapy that can deliver drugs with a high level of selectivity towards a specific cell type, reducing off-target effects and increasing the therapeutic window compared to

Senescent Cell Derived Artificial Vesicle‐Based Senolytic Sonovaccine Platform with Augmented Lymph Node Delivery and Antigen Cross‐Presentation Efficacy

open access: yesAdvanced Science, EarlyView.
A senolytic sonovaccine platform (SenoVac) is developed, in which senescent cell‐derived vesicles serve as broad senescent cell antigen reservior. The “2‐step” click chemistry strategy for effective lymph node delivery, and ultrasound‐triggered endosomal escape to boost cross‐presentation, ensures efficient senescent cell clearance and disease ...

SuFEx‐Enabled Reprogramming of Flavonoids for Selective α‐Glucosidase Covalent Inhibition

open access: yesAdvanced Science, EarlyView.
Selective inhibition of intestinal α‐glucosidase remains limited by poor enzyme specificity and off‐target metabolic effects. Here, SuFEx click chemistry is used to reprogram natural flavonoids into covalent inhibitors with enhanced α‐glucosidase selectivity and controlled reactivity. This strategy enables effective regulation of carbohydrate digestion

PI3Kδ activation, IL-6 overexpression, and CD37 loss cause resistance to naratuximab emtansine in lymphomas

open access: yesBlood Advances
: CD37-directed antibody and cellular-based approaches have shown preclinical and promising early clinical activity. Naratuximab emtansine (Debio 1562; IMGN529) is an antibody-drug conjugate (ADC) incorporating an anti-CD37 monoclonal antibody conjugated

Antibody-Drug Conjugates for Cancer Therapy: Chemistry to Clinical Implications

open access: yesPharmaceuticals, 2018
Chemotherapy is one of the major therapeutic options for cancer treatment. Chemotherapy is often associated with a low therapeutic window due to its poor specificity towards tumor cells/tissues.

CCL3 and IL‐7 Synergistically Enhance CAR‐T Efficacy in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Synergistic delivery of CCL3 and IL‐7 by CAR‐T cells overcomes the immunosuppressive tumor microenvironment by enhancing cell infiltration, survival, and local immune reprogramming. This also establishes positive feedback that amplifies anti‐tumor activity and memory responses in both CAR‐T and endogenous T cells, presenting a robust therapeutic ...

A DNA-Interacting Payload Designed to Eliminate Cross-Linking Improves the Therapeutic Index of Antibody–Drug Conjugates (ADCs) [PDF]

open access: yes, 2018
Tumor-selective delivery of cytotoxic agents in the form of antibody–drug conjugates (ADCs) is now a clinically validated approach for cancer treatment. In an attempt to improve the clinical success rate of ADCs, emphasis has been recently placed on the ...

Tumor‐Specific Delivery of CD28 siRNA via Lyso‐PC C‐16 Modified Lipid Nanoparticles Overcomes Anti‐PD‐1 Resistance by Remodeling Tumor Microenvironment

open access: yesAdvanced Science, EarlyView.
This study develops 16:0 LPC‐modified lipid nanoparticles (LPC‐LNPs) with cancer cell specificity by exploiting altered tumor lipid metabolism. LPC‐LNPs encapsulating Cd28 small interfering RNA (LPC‐LNP‐Cd28) knock down cancer cell CD28 without affecting T cells, inflame the tumor microenvironment, and overcome anti‐PD‐1 resistance.
