Cystine (Cys2) deprivation in pancreatic cancer cells induces oxidative stress that destabilizes cytosolic iron–sulfur cluster (ISC) proteins, triggering an iron‐regulatory protein (IRP)‐mediated iron‐starvation response (ISR). This leads to increased iron uptake (via TFRC), an expanded labile iron pool, and ferroptosis.

Saquayamycin B1 (SQ‐B1), an antibiotic isolated from Streptomyces sp. I072, triggers cell death in different melanoma cell lines, including chemoresistant SK‐MEL‐5. SQ‐B1 elevates O2•‐ and NO• on both sides of the membrane and blocks glutathione transferase (GSTP‐1) and glutathione reductase (GSR), destabilizing the GSH/GSSG antioxidant system.
