ABSTRACT Proteolysis‐targeting chimeras (PROTACs) are heterobifunctional molecules that hijack the ubiquitin‐proteasome system to drive catalytic, sub‐stoichiometric degradation of disease‐associated proteins, offering a mechanistic advantage over occupancy‐driven inhibitors and access to ‘undruggable’ targets.
