An isoxazole strategy for the synthesis of curcumin and demethoxycurcumin
Curcumin and demethoxycurcumin were synthesized via an isoxazole-masking strategy. A 3,5-disubstituted isoxazole, obtained by [3 + 2] cycloaddition of a nitrile oxide and an alkyne, underwent mild reductive N-O cleavage to reveal the β-diketone core in good yield.

Selective Anti‐Inflammatory Actions of Curcumin on Cytokine Pathways in Major Depressive Disorder
Curcumin partially attenuates IRS/M1/Th1 immune activation in MDD patients ex vivo, but fails to normalize it. Paradoxically, it upregulates immune profiles in healthy controls. Its immunomodulatory effects are state‐dependent, suggesting cautious use in non‐depressed individuals.
