A novel USP4 inhibitor that suppresses colorectal cancer stemness by promoting β-catenin and Twist1 degradation

open access: yesJournal of Translational Medicine
Background The high mortality rate of metastatic colorectal cancer (CRC) is primarily attributed to resistance to chemotherapy, where cancer stem cells (CSCs) play a crucial role.

Phase Separation of TRIM21 Modulates PTPN14 Stability to Drive Flow‐Dependent Endothelial Activation and Atherogenesis

open access: yesAdvanced Science, EarlyView.
Disturbed flow promotes the formation of TRIM21‐rich biomolecular droplets, which concentrate TRIM21 and PTPN14 and facilitate their SPRY‐FERM interaction (illustrated by the TRIM21 D355‐PTPN14 R132 salt bridge). This condensate‐driven proximity enables TRIM21 to catalyze K48‐linked polyubiquitination of PTPN14 at lysine 956, leading to proteasome ...

DataSheet_1_Modified activities of macrophages’ deubiquitinating enzymes after Francisella infection.docx

open access: yes, 2023
Francisella tularensis influences several host molecular/signaling pathways during infection. Ubiquitination and deubiquitination are among the most important regulatory mechanisms and respectively occur through attachment or removal of the ubiquitin ...

Selective Modulation of OTUB1 Noncanonical Function via a bioPhosTAC Strategy

open access: yesAdvanced Science, EarlyView.
This work positions the versatile performance of the peptide‐based bioPhosTAC platform for dissecting phosphorylation‐dependent biology and expanding the scope of induced‐proximity technologies. We demonstrated that selective manipulation of a tyrosine phosphorylation site is sufficient to propagate coordinated cellular consequences.

Regulation of proteolysis by human deubiquitinating enzymes

open access: yesBiochimica et Biophysica Acta (BBA) - Molecular Cell Research, 2014
The post-translational attachment of one or several ubiquitin molecules to a protein generates a variety of targeting signals that are used in many different ways in the cell. Ubiquitination can alter the activity, localization, protein-protein interactions or stability of the targeted protein.

GSTM4 Suppresses Prostate Cancer Progression by Targeting TRIM27‐mediated ACLY Ubiquitination and Lipid Metabolism

open access: yesAdvanced Science, EarlyView.
GSTM4 enhances the interaction between ACLY and the E3 ubiquitin ligase TRIM27, thereby increasing K48‐linked ubiquitination at the K554 site of ACLY and promoting its degradation. Consequently, GSTM4 regulates lipid synthesis, as well as the proliferation and metastasis of PCa cells, by modulating ACLY stability.

Immunoexpression of tumor suppressor protein p53 and deubiquitinating enzymes in oral squamous cell carcinoma

open access: yes, 2022
The ubiquitin-proteasome system is regulated by deubiquitinating enzymes (DUBs), which include the complex Ubiquitin-specific protease 1 (USP1) and WD40 repeat-containing protein 48 (WDR48).

Preliminary study of the enzyme ubiquitin carboxylterminal hydrolase 14 (UBP6) in Giardia intestinalis: structural bioinformatic analysis and transcriptional profile during encystation

open access: yesRevista Colombiana de Química, 2014
This paper presents a combined approach with two aims. The first is to analyze the reported sequence of the enzyme ubiquitin carboxyl-terminal hydrolase 14 of Giardia intestinalis (UBP6) through computational methods to find components related with its ...

Deubiquitination of Vangl by USP6 and USP32 Regulates Planar Cell Polarity Signaling

open access: yesAdvanced Science, EarlyView.
Compartment‐specific deubiquitination controls Vangl dosage and planar cell polarity signaling. USP6 and USP32 regulate distinct subcellular pools of Vangl by removing distinct ubiquitin modifications from Vangl proteins. This regulatory mechanism safeguards PCP‐dependent embryonic morphogenesis, while aberrant USP32‐dependent stabilization of VANGL ...
