A method is described to determine the position of fatty acids on lecithin. Purified egg lecithin is enzymatically hydrolyzed to the corresponding mixture of α,β-diglycerides with lecithinase D.

Simple Method for Derivatization of Monoglycerides and Diglycerides
Abstract A method has been developed for derivatization of monoglycerides and diglycerides to their propyl esters by using propionic anhydride in the presence of pyridine. The derivatives are volatile and thus can be separated and identified by gas chromatography (GC). Completion of the reaction is monitored by thin-layer chromatography.

Abstract CDP-diglyceride synthesized by chemical procedures is an efficient donor of phosphatidyl residues for the enzymatic synthesis of phospholipids in cell-free extracts of Escherichia coli. However, prior to the present work, CDP-diglyceride had never been isolated from living cells of E. coli or any other organism.
