Pre‐treatment DPYD and UGT1A1 genotyping is increasingly used to prevent fluoropyrimidine‐ and irinotecan‐related toxicity, but variant‐specific real‐world effects remain unclear. In an unselected cohort of cancer patients with actionable genotypes, genotype‐driven dosing improved safety while preserving treatment exposure in high‐risk DPYD c.1905+1G>A

Case report: 5-Fluorouracil treatment in patient with an important partial DPD deficiency
Esophageal cancer is a cancer with poor prognosis and the standard 1st line treatment for metastatic or recurrent EC is systemic chemotherapy with doublet chemotherapy based on platinum and 5-fluorouracil (5-FU). However, 5-FU could be a source of severe

ABSTRACT β‐Aminoisobutyric acid (BAIBA) is an exercise‐responsive myokine/metabolite released by skeletal muscle and has attracted increasing interest as a potential exercise mimetic and therapeutic candidate in preclinical research. BAIBA acts locally in skeletal muscle and can also reach distal organs through the circulation, contributing to the ...

EXOSC10, an essential nuclear RNA exosome‐associated 3′‐5′ exoribonuclease, is inhibited by the anticancer drug 5‐fluorouracil (5‐FU), and EXOSC10 depletion increases 5‐FU sensitivity. The colon‐cancer variant EXOSC10S402T, located in a proteolysis motif, is stable and nuclear but nonfunctional in vivo.

Analysis of the dihydropyrimidine dehydrogenase polymorphism in a British population [PDF]
Subjects with low or absent dihydropyrimidine dehydrogenase activity (DPD) are at risk of excessive toxicity or death when undergoing fluoropyrimidine chemotherapy. The DPD polymorphism has not been well characterized in the general population and the frequency of the enzyme deficiency is not known.

Thermal and photochemical reactions of dihydrodiazines [PDF]
This thesis describes the results of an investigation into the thermal and photochemical reactivity of dihydrodiazines. In order to prepare the title compounds the diazines and some phenyldiazines are treated with phenyllithium in ether, yielding adducts ...

Characterization of the Human Dihydropyrimidine Dehydrogenase Gene
Dihydropyrimidine dehydrogenase (DPD) catabolizes endogenous pyrimidines and pyrimidine-based antimetabolite drugs. A deficiency in human DPD is associated with congenital thymine-uraciluria in pediatric patients and severe 5-fluorouracil toxicity in cancer patients. The dihydropyrimidine dehydrogenase gene (DPYD) was isolated, and its physical map and
