Cigarette smoke accelerates the aging of immune cells in the lung. By combining human single‐cell sequencing, cell culture, and mouse models, the authors show that the protein GEM drives mitochondrial damage and senescence in alveolar macrophages by suppressing SIRT3.

Understanding DNA Damage Response and DNA Repair in Multiple Myeloma
Simple Summary Multiple myeloma (MM), a malignant plasma cell disorder, is characterized by abnormal DNA damage response (DDR). MM cells adapt over time via gene mutations or chromosomal aberrations, including changes to DDR and DNA repair genes ...

The RNA Processing Factor Y14 Participates in DNA Damage Response and Repair
Summary: DNA repair deficiency leads to genome instability and hence human disease. Depletion of the RNA processing factor Y14/RBM8A in cultured cells or Rbm8a haplodeficiency in the developing mouse cortex results in the accumulation of DNA damage.

Integrative multi‐omics analysis delineates a mitochondrial–immune axis governing neoadjuvant chemotherapy response in high‐grade serous ovarian cancer. Immune‐active tumors exhibit enhanced B‐cell infiltration and favorable sensitivity, whereas metabolically rewired tumors display oxidative phosphorylation dependency and resistance.

Molecular and Cellular Hallmarks of Age‐Related Vestibular Hair Cell Degeneration
This study utilizes single‐cell RNA‐seq transcriptomes, advanced imaging, and electrophysiology to examine universal and cell‐type‐specific aging signatures of vestibular hair cells. The study shows that impaired hair bundle function is a key driver of age‐related vestibular dysfunction.

NKX3.1 contributes to S phase entry and regulates DNA damage response (DDR) in prostate cancer cell lines [PDF]
NKX3.1 is an androgen-regulated homeobox gene that encodes a tissue-restricted transcription factor, which plays an important role in the differentiation of the prostate epithelium. Thus, the role of NKX3.1 as a functional topoisomerase I activity enhancer in cell cycle regulation and the DNA damage response (DDR) was explored in prostate cancer cell ...

Penfluridol Triggers GSDME‐Mediated Immunogenic Pyroptosis to Potentiate Antitumor Immunotherapy
A high‐throughput screen of FDA‐approved antipsychotics identifies penfluridol as a potent pyroptosis inducer acting via direct TTI1 inhibition. This triggers TNFA‐NFKB signaling and caspase‐8/‐3‐dependent GSDME cleavage. The compound stimulates antitumor immunity alone and synergizes with anti‐PD‐1 therapy, while low TTI1 expression emerges as a ...

Monitoring the Activation of the DNA Damage Response Pathway in a 3D Spheroid Model. [PDF]
Monitoring the DNA-Damage Response (DDR) activated pathway in multicellular tumor spheroid models is an important challenge as these 3D models have demonstrated their major relevance in pharmacological evaluation.
