Degradation mechanism of the von Willebrand factor A2 domain by nattokinase

Nattokinase, a natto‐derived protease, exhibits potent antithrombotic effects. This study demonstrates that nattokinase directly cleaves the von Willebrand factor (vWF) A2 domain in vitro. Unlike the native regulator ADAMTS13, nattokinase degrades folded vWF independently of shear stress.

Structural insights and therapeutic targets in Acinetobacter baumannii capsule biosynthesis

Hypervirulent KL49 A. baumannii's capsular polysaccharide contains the nonulosonic acid 8‐epi‐Leg5,7Ac2, synthesized by epimerization via ElaA, ElaB, and ElaC. Crystal structures of ElaA, ElaB, and ElaC reveal their role in CMP‐Leg5,7Ac2 synthesis and regioselective C8 epimerization.

近年,ヒト肝臓において非常に多くのトランスポーターが同定・機能解析されるにつれて,トランスポーターの遺伝子多型や薬物間相互作用による機能変動が薬物動態に与える影響を明らかにするための臨床研究も続々と報告されてきている.それに伴い,異物解毒システムの中でのトランスポーターの重要性が広く認知されてきた.代謝によって消失すると考えられてきた薬物の中にも肝取り込みトランスポーターの基質が含まれていることが明らかとなり,取り込みトランスポーターの機能変動が薬物動態の変化につながる事例が複数報告されてきている.本稿では,ヒト肝臓に発現する主な薬物トランスポーターを紹介すると共に,これらの遺伝子多型・薬物間相互作用が薬物動態や薬効・副作用に与える影響について概説することを目指した.
