Electron Cryo-Tomography Reveals the Caulobacter vibrioides Tight Adherence Pilus Architecture
ABSTRACT The type IV filament superfamily is a widespread group of molecular machines involved in natural transformation, motility, adhesion, nutrient uptake, and secretion of a wide spectrum of protein substrates. The gram-negative bacterium Caulobacter vibrioides expresses the ...

A HER3‐targeted sonogenetic lipid nanoparticle platform is developed for triple‐negative breast cancer therapy. An AI‐designed miniprotein inhibits HER3 signaling, while TRPV4 and NFAT‐responsive IL‐15 plasmids enable ultrasound‐controlled calcium signaling and cytokine expression. The system induces Ca2+ overload, ROS generation, mitochondrial damage,

Hederagenin is identified as a GABAA1‐targeting natural compound that suppresses morphine‐associated reward by restraining Ca2+–CaMK–cAMP–CREB signaling in ventral tegmental dopaminergic neurons. RVG29‐modified liposomal delivery further enhances brain accumulation and behavioral efficacy, providing a mechanistically defined strategy for natural ...

An orally administered, drug‐free hyaluronic acid nanoassembly, uhNAs‐OA, accumulates in the inflamed colon and is preferentially internalized by M1‐like macrophages via HA‐CD44 interactions. uhNAs‐OA scavenges excess ROS, suppresses NF‐κB signaling and remodels the colonic immune landscape, resulting in robust therapeutic efficacy in acute, chronic ...

An injectable and self‐reinforcing RSF‐based piezoelectric hydrogel conforms to irregular bone defects and progressively strengthens in situ, overcoming the trade‐off between injectability and mechanical stability. Mechanical loading generates bioelectrical cues that promote P300‐mediated H3K9 acetylation and upregulate PTH1R expression, thereby ...

Here, a PDL1 inhibitor is employed as a fifth component to modify lipid nanoparticles (LNPs). PDL1 inhibitor‐loaded LNPs show enhanced lymph node targeting, resulting in greater lymph node expansion, immune cell infiltration, and improved mRNA vaccine efficacy. This fifth component LNP could be a novel platform for mRNA vaccines.

View Tomo: Context-aware targeting and analysis in electron cryo-tomography
Abstract Electron cryo-tomography (cryoET) resolves cellular structure in three dimensions, yet region selection is still typically based on two-dimensional projection images. Here, we introduce View Tomo, a workflow for rapid acquisition of low-magnification tomograms that enables screening, targeting and analysis in

A deep‐learning‐designed binder is fused to a combinatorial degron to create a binder‐degron chimera (bdC) that recruits the CUL2‐RING ubiquitin ligase complex. Encapsulation in a prostate‐cancer‐targeted, CD47‐camouflaged nanocarrier enables tumor‐specific in situ bdC expression, triggering GPX4 degradation and ferroptosis, thereby establishing a ...
