Inhibition of HMGA2 Leads to Reduced Cell Proliferation and Increased Apoptosis in Human Embryonal Carcinoma Cell Lines

open access: yesAndrology, EarlyView.
ABSTRACT Background The most prevalent solid tumors in young men are testicular germ cell tumors (TGCTs), and embryonal carcinoma is the most common subtype among non‐seminomatous germ cell tumors (NSGCTs). Despite the excellent cure rates of cisplatin‐based chemotherapy, resistance develops in 15%–30% of patients with metastatic cancer, which results ...

Testis‐Enriched F‐Box Protein FBXO39 Is Important for Spermiogenesis and Male Fertility in Mice

open access: yesAndrology, EarlyView.
ABSTRACT Background The SCF (Skp–Cullin–F‐box) complex is a major class of E3 ubiquitin ligases. F‐box proteins constitute the SCF complex and play a critical role in recognizing substrates for ubiquitination. In mice, several F‐box proteins, including FBXO36 and FBXO39, are predominantly expressed in testes.

Focal therapy using a novel cooled laser device for prostate cancer: early trial results

open access: yesBJU International, EarlyView.
Objectives To determine the short‐term oncological results and safety of the ProFocal Laser Therapy for Prostate Tissue Ablation (PFLT‐PC) trial, the first phase II clinical trial of ProFocal® (Medlogical Innovations, Sydney, Australia), a novel, cooled laser focal therapy (cLFT) device for prostate cancer (PCa) treatment. Patients and Methods Men with

Intermediate clinical endpoints as surrogates for overall survival after salvage prostatectomy

open access: yesBJU International, EarlyView.
Objectives To evaluate biochemical recurrence‐free survival (BRFS) and metastasis‐free survival (MFS) as potential intermediate clinical endpoints (ICEs) for overall survival (OS) in patients undergoing salvage radical prostatectomy (sRP). Patients and Methods Evaluable patients were selected from a retrospective dataset, resulting in a cohort of 879 ...

PDE4D and PDE3B orchestrate distinct cAMP microdomains in 3T3‐L1 adipocytes

open access: yesBritish Journal of Pharmacology, EarlyView.
Basal conditions: •Ins/PDE3B lowers cytoplasmic cAMP (cyt‐cAMP) without affecting plasma membrane cAMP (pm‐cAMP). •Insulin decreases lipid droplet cAMP (LD‐cAMP) independent of PDE3B. •FGF1/PDE4D modestly reduces both cyt‐ and pm‐cAMP, while PDE4D alone can modulate LD‐cAMP. ISO stimulation: •Ins/PDE3B has minimal impact on cyt‐cAMP.

A novel selective stabilizer of the ryanodine receptor 2 prevents stress‐induced ventricular arrhythmias without impairing cardiac function

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract Background and Purpose Aberrant activation of type 2 ryanodine receptors (RyR2) causes lethal arrhythmias, such as catecholaminergic polymorphic ventricular tachycardia (CPVT). Developing drugs that suppress RyR2 hyperactivation may be key to novel arrhythmia treatments.
