Screening 166 FDA‐approved anticancer drugs identifies the aromatase inhibitor Exemestane as a synergistic partner of PARP inhibitor Olaparib in BRCA‐proficient triple‐negative breast cancer. Exemestane induces ROS‐mediated replication stress, enhancing DNA damage and apoptosis alongside Olaparib.

Flavonoids are plant bioactives that are recognized as hormone-like polyphenols because of their similarity to the endogenous sex steroids 17β-estradiol and testosterone, and to their estrogen- and androgen-like activity. Most efforts to verify flavonoid

Estrogen Receptors and Melanoma: A Review [PDF]
In the last three decades cutaneous melanoma has been widely investigated as a steroid hormone-sensitive cancer. Following this hypothesis, many epidemiological studies have investigated the relationship between estrogens and melanoma. No evidence to date has supported this association due to the great complexity of genetic, external and environmental ...

Development of 4T1 breast cancer mouse model system for preclinical carbonic anhydrase IX studies
Carbonic anhydrase IX (CAIX) is a well‐recognised therapeutic target and prognostic biomarker in cancer. We developed and characterised a robust murine breast cancer model system that is suitable for CAIX studies in vitro and in vivo—it comprises both CAIX‐positive and CAIX‐negative controls and provides a solid platform for the comprehensive ...

Caloric restriction that extends lifespan induces the expression of PGC‐1α and MIPEP in white adipose tissue. In this study, co‐overexpression of Pgc‐1α and Mipep upregulated the gene expression of PHOSPHO1. These findings provide new insights into mitochondria‐related mechanisms underlying the effects of caloric restriction in adipocytes.

Palmitoylation of estrogen receptors is essential for neuronal membrane signaling.
In addition to activating nuclear estrogen receptor signaling, 17β-estradiol can also regulate neuronal function via surface membrane receptors. In various brain regions, these actions are mediated by the direct association of estrogen receptors (ERs ...

Molecular mechanism of estrogen–estrogen receptor signaling [PDF]
Abstract17β‐Estradiol (E2), as the main circulating estrogen hormone, regulates many tissue and organ functions in physiology. The effects of E2 on cells are mediated by the transcription factors and estrogen receptor (ER)α and ERβ that are encoded by distinct genes.

ABSTRACT Objective The Apolipoprotein (APOE) ε4 allele is the strongest genetic risk factor for late‐onset Alzheimer's disease (AD); however, many ε4 carriers remain cognitively intact into old age. Leveraging plasma neuron‐derived extracellular vesicles (NDEVs), we sought to identify biomarkers of cognitive resilience and their interplay with APOE ...
