Design, Synthesis, and Biological Evaluation of Pyrazolyl Thioureas (PTUs) as SIRT1 and SIRT2 Inhibitors

Pyrazolyl thioureas were designed and synthesized as sirtuin 1 (SIRT1) and sirtuin 2 (SIRT2) inhibitors. 6a (IC50 (SIRT1) = 5.3 µM; IC50 (SIRT2) = 2.2 µM) and 7f (IC50 (SIRT1) = 12.4 µM; IC50 (SIRT2) = 13.6 µM) showed micromolar inhibition on target enzymes and HepG2‐based assays confirmed a SIRT2‐mediated effect on tubulin acetylation.
