Reconstitution of apo-DT-diaphorase with flavin-adenine dinucleotide.

The binding of flavin-adenine dinucleotide (FAD) in DT-diaphorase was studies. Incubation with 4M guanidine-HCl at 0°C rapidly dissociated FAD from the enzyme, and incubation of the apo-enzyme with FAD at 37°C restored maximally about 50% (for menadione-reducing activity), or 330% (for ferricyanide-reducing activity) of the initial activity.

Photochemistry of CryB from Rhodobacter sphaeroides

Elucidation of the photochemistry of CryB from Rhodobacter sphaeroides was achieved from the nanosecond to second timescale. A fast (<5 ns) deprotonation of the terminal electron donor Trp338 and the involvement of Tyr337 in photoreduction have been revealed.

FLAVIN-ADENINE DINUCLEOTIDE-SYNTHESIZING ENZYME IN EREMOTHECIUM ASHBYII

The existence of an enzyme catalyzing FAD synthesis from FMN and ATP was proved in mycelium of Er. ashbyii. The enzyme requires Mg2+ or Mn2+ for its full activity. The optimum pH is 8.0. When riboflavin is added to the reaction mixture in place of FMN, FAD synthesis occurs also, but not so much as in the case of FMN.
