Guidelines for the use of flow cytometry and cell sorting in immunological studies (second edition)

open access: yesEuropean Journal of Immunology, 2019
These guidelines are a consensus work of a considerable number of members of the immunology and flow cytometry community. They provide the theory and key practical aspects of flow cytometry enabling immunologists to avoid the common errors that often ...

Monitoring CAR T‐cells using flow cytometry

open access: yesCytometry. Part B, Clinical cytometry, 2020
Chimeric antigen receptor (CAR) T‐cell therapy is considered as a major scientific breakthrough in cancer immunotherapy. The success of adoptive CAR T‐cell therapy for cancer has inspired researchers to expand indications into the area of solid tumors ...

β‐TrCP overexpression enhances cisplatin sensitivity by depleting BRCA1

open access: yesMolecular Oncology, EarlyView.
Low levels of β‐TrCP (Panel A) allow the accumulation of BRCA1 and CtIP, which facilitate the repair of cisplatin‐induced DNA damage via homologous recombination (HR) and promote tumor cell survival. In contrast, high β‐TrCP expression (Panel B) leads to BRCA1 and CtIP degradation, impairing HR repair, resulting in persistent DNA damage and apoptosis ...

Harmonization of Flow Cytometric Minimal Residual Disease Assessment in Multiple Myeloma in Centers of Polish Myeloma Consortium

open access: yesDiagnostics, 2021
Minimal residual disease (MRD) status is now considered as one of the most relevant prognostic factors in multiple myeloma (MM) while MRD negativity became an important endpoint in clinical trials.

Validation of CyTOF Against Flow Cytometry for Immunological Studies and Monitoring of Human Cancer Clinical Trials

open access: yesFrontiers in Oncology, 2019
Flow cytometry is a widely applied approach for exploratory immune profiling and biomarker discovery in cancer and other diseases. However, flow cytometry is limited by the number of parameters that can be simultaneously analyzed, severely restricting ...

Reproducibility of Flow Cytometry Through Standardization: Opportunities and Challenges

open access: yesCytometry Part A, 2020
There is an agreement in the field that interlaboratory reproducibility of flow cytometry measurements as well as the whole studies might be improved by a consensual use of methodological approach.

Modeling hepatic fibrosis in TP53 knockout iPSC‐derived human liver organoids

open access: yesMolecular Oncology, EarlyView.
This study developed iPSC‐derived human liver organoids with TP53 gene knockout to model human liver fibrosis. These organoids showed elevated myofibroblast activation, early disease markers, and advanced fibrotic hallmarks. The use of profibrotic differentiation medium further amplified the fibrotic signature seen in the organoids.

Comprehensive Phenotyping of T Cells Using Flow Cytometry

open access: yesCytometry Part A, 2019
The T cell compartment can form a powerful defense against extrinsic (e.g., pathogens) and intrinsic danger (e.g., malignant cells). At the same time, specific subsets of T cells control this process to keep the immune system in check and prevent ...

PYCR1 inhibition in bone marrow stromal cells enhances bortezomib sensitivity in multiple myeloma cells by altering their metabolism

open access: yesMolecular Oncology, EarlyView.
This study investigated how PYCR1 inhibition in bone marrow stromal cells (BMSCs) indirectly affects multiple myeloma (MM) cell metabolism and viability. Culturing MM cells in conditioned medium from PYCR1‐silenced BMSCs impaired oxidative phosphorylation and increased sensitivity to bortezomib.

The utility of flow cytometry for the detection of tumor cells in cerebrospinal fluid of patients with acute leukemia

open access: yesUniversitas Scientiarum, 2022
Central nervous system infiltration by acute leukemia is a poor prognosis variable, and conventional cytology is the gold standard for its diagnosis; the technique is highly specific but not sensitive.
