Invariant natural killer T (iNKT) cells are T lymphocytes that behave similarly to cells of the innate immune system. The glycolipid alpha-galactosylceramide (alpha-GalCer) is a potent and specific activator of mouse and human iNKT cells and has been used in cancer clinical trials to drive NKT cell-mediated immune responses.

The RNA‐ and DNA‐Binding Protein Y‐Box Binding Protein 1 (YB‐1) Regulates iNKT Cell Development
YB‐1 is essential for invariant NKT cell development, integrating TCR, co‐stimulatory, and IL‐15 signaling to control maturation, subset differentiation, and survival. Its loss at the double‐positive thymocyte stage causes ∼80% iNKT cell depletion, maturation arrest, impaired cytokine production, and increased apoptosis.

Synthesis of Galactosylceramide and Glucosylceramide by Mouse Kidney Preparations
A particulate fraction was prepared from kidneys, lyophilized, and coated with ceramide. After incubation with UDP-[14C]Gal and appropriate cofactors, the major radioactive component in the lipid fraction was galactosylceramide (Gal-Cer). If UDP-Glc was substituted for UDP-Gal, the product was glucosylceramide (Glc-Cer).
