Preparative scale isolation of sphingosine.
A high-yield method is described for the preparation of sphingenine from galactosylceramide. Two successive cleavage reactions are used, alkaline butanol to form galactosylsphingosine, then acidic acetonitrile to form the sphingenine.

Summary: Doxorubicin (DOX)-induced cardiomyopathy has poor prognosis, and myocardial inflammation is intimately involved in its pathophysiology. The role of invariant natural killer T (iNKT) cells has not been fully determined in this disease.

Mixed Signals: T Cells as Architects of IgE Immunity
ABSTRACT Food allergen‐specific IgE can cause significant pathology, yet the pathways that generate pathogenic, high‐affinity IgE remain incompletely understood. Increasing evidence suggests that IgE responses arise from the integration of multiple, and sometimes opposing, T cell–derived cues.

Abnormalities of glycosphingolipids in mucopolysaccharidosis type III B.
Glycosphingolipids from brain, liver, and spleen of a patient with mucopolysaccharidosis type III B were quantitatively analyzed. Neutral glycosphingolipids containing glucosylceramide, lactosylceramide, globotriaosylceramide, globotetraosylceramide, and

Suppression of eosinophilic airway inflammation by treatment with α‐galactosylceramide [PDF]
AbstractTo clarify the essential role of NKT cells in allergy, we investigated the contribution of NKT cells to the pathogenesis of eosinophilic airway inflammation using α‐galactosylceramide (α‐GalCer), a selective ligand for NKT cells. Although continuous administration of α‐GalCer during ovalbumin (OVA) sensitization increased OVA‐specific IgE ...

This graphical abstract illustrates the gut microenvironment disparities between breastfed and formula‐fed infants at 6 months of age. Through multi‐omics analysis, breastfeeding was shown to establish a distinct ecological imprinting characterized by favorable microbial shifts and down‐regulated pro‐inflammatory arachidonic acid metabolism, thereby ...

Leishmania Extracellular Vesicles as a Preventive Vaccine Platform Against Leishmaniasis
The potential of Leishmania‐derived extracellular vesicles (EVs) as a vaccine platform was investigated, emphasizing an affordable, GMP‐compliant production and purification method. Immunization with EV vaccines, either unadjuvanted or combined with α‐galactosylceramide (αGC), induced robust humoral and cellular immune responses, conferring protection ...

Heparan Sulfate Proteoglycans in Immunity
ABSTRACT In immune cells, heparin was first identified in mast cells in the 1940s, followed by the discovery of heparan sulfate in lymphocytes and granulocytes during the 1970s and 1980s. Subsequent decades of research have revealed how heparan sulfate and heparin, conjugated to core proteins as heparan sulfate proteoglycans (HSPGs), act as versatile ...

Biological Roles Played by Sphingolipids in Dimorphic and Filamentous Fungi
Filamentous and dimorphic fungi cause invasive mycoses associated with high mortality rates. Among the fungal determinants involved in the establishment of infection, glycosphingolipids (GSLs) have gained increased interest in the last few decades.
