The nicotinamide hypothesis revisited—plant defense signaling integrating PARP, nicotinamide, nicotinic acid, epigenetics, and glutathione

open access: yesFEBS Letters, EarlyView.
Nicotinamide (NIC) and nicotinic acid (NIA) are proposed as stress signaling compounds in plants. Oxidative stress may lead to single strand breaks (SSB) in DNA, which activate poly(ADP‐ribose) polymerase (PARP). NIC and NIA are then formed from NAD. NIC and NIA can promote epigenetic changes leading to the expression of defense genes specific for the ...

Induction of RNAi Core Machinery’s Gene Expression by Exogenous dsRNA and the Effects of Pre-exposure to dsRNA on the Gene Silencing Efficiency in the Pea Aphid (Acyrthosiphon pisum)

open access: yesFrontiers in Physiology, 2019
The pea aphid, Acyrthosiphon pisum, is an important agricultural pest and biological model organism, and RNA interference (RNAi) is an important tool for functional genomics and for insect pest management. However, the efficiency of RNAi in pea aphids is

Bidirectional transcription directs both transcriptional gene activation and suppression in human cells. [PDF]

open access: yesPLoS Genetics, 2008
Small RNAs targeted to gene promoters in human cells have been shown to modulate both transcriptional gene suppression and activation. However, the mechanism involved in transcriptional activation has remained poorly defined, and an endogenous RNA ...

From lactation to malignancy: A comparison between healthy and cancerous breast gland at single‐cell resolution reveals new issues for tumorigenesis

open access: yesFEBS Letters, EarlyView.
Single‐cell RNA sequencing reveals an opposite role of SLPI in basal tumors based on metastatic spread, along with shared activation of specific regulons in cancer cells and mature luminal lactocytes, as well as downregulation of MALAT1 and NEAT1 in the latter.

Development and application of a virus-induced gene silencing protocol for the study of gene function in narrow-leafed lupin

open access: yesPlant Methods, 2021
Background Lupins are promising protein crops with an increasing amount of genomic and transcriptomic resources. The new resources facilitate the in silico identification of candidate genes controlling important agronomic traits.

Transfer RNA-derived small RNAs in the cancer transcriptome [PDF]

open access: yes, 2016
The cellular lifetime includes stages such as differentiation, proliferation, division, senescence and apoptosis. These stages are driven by a strictly ordered process of transcription dynamics.

Cancer cell death induced by the NAD antimetabolite Vacor discloses the antitumor potential of SARM1

open access: yesFEBS Letters, EarlyView.
Vacor, a compound converted into the toxic metabolite Vacor adenine dinucleotide (VAD) by the nicotinamide salvage pathway enzymes NAMPT and NMNAT2, exhibits antitumor activity by inducing rapid and complete NAD depletion. We report that Vacor toxicity is limited to cell lines expressing high levels of SARM1, a NAD glycohydrolase.

Evolutionary Genetics: The silence of the genes [PDF]

open access: yesCurrent Biology, 1995
A low mutation rate is required for the evolution of large genomes. But is the repression of inadvertent gene expression also important, and is the evolution of complexity limited by the efficiency of noise reduction?

Intramyocardial gene silencing by interfering RNA [PDF]

open access: yes, 2017
RNAi is a widely used methodology for gene silencing. The action mechanism of siRNA molecules has been well studiedin recent years, and the technique has been optimized in terms of safety and effectiveness. Cardiovascular diseases havea high incidence in

The MBD7 complex promotes expression of methylated transgenes without significantly altering their methylation status. [PDF]

open access: yes, 2017
DNA methylation is associated with gene silencing in eukaryotic organisms. Although pathways controlling the establishment, maintenance and removal of DNA methylation are known, relatively little is understood about how DNA methylation influences gene ...
