The mitochondrial outer membrane iron–sulphur ([Fe‐S]) protein mitoNEET is a target of the type‐2 diabetes drug pioglitazone. Its unknown molecular function is linked to respiratory complex activity and mitochondrial function. We discovered that O2 protects the mitoNEET [2Fe‐2S] cluster against NO oxidation and desensitization towards reduction by H2S.

Effective gene targeting in rabbits using RNA-guided Cas9 nucleases.
Dear Editor, Recently, zinc finger nuclease, transcription activator-like effector nuclease, and RNA-guided Cas9 endonuclease (Cas9) have emerged as powerful means for genome editing (Conklin, 2013; Gaj et al., 2013).

Characterization of fungal carbonyl sulfide hydrolase belonging to clade D β‐carbonic anhydrase
Here, we performed a functional analysis of the fungal enzymes belonging to clade D of the β‐class carbonic anhydrase family (β‐D‐CA). The β‐D‐CAs in the basidiomycete Gloeophyllum trabeum and the ascomycete Trichoderma harzianum showed very low activity in the hydration of CO2 but exhibited high activity in the hydrolysis of carbonyl sulfide (COS ...

Screening for the optimal siRNA targeting a novel gene (HA117) and construction and evaluation of a derivative recombinant adenovirus [PDF]
We found a novel gene named as HA117 in our previous research. At this study, we screened for an optimal siRNA targeting the novel gene HA117 using the pSOS-HUS method, verified the results of pSOS-HUS siRNA screening for optimal affinity for the target ...

Life cycle studies of the hexose transporter of Plasmodium species and genetic validation of their essentiality [PDF]
A Plasmodium falciparum hexose transporter (PfHT) has previously been shown to be a facilitative glucose and fructose transporter. Its expression in Xenopus laevis oocytes and the use of a glucose analogue inhibitor permitted chemical validation of PfHT ...

Machine learning discriminates a movement disorder in a zebrafish model of Parkinson's disease
Animal models of human disease provide an in vivo system that can reveal molecular mechanisms by which mutations cause pathology, and, moreover, have the potential to provide a valuable tool for drug development. Here, we have developed a zebrafish model

We explore 5HT2A receptor activation, a key target for psychedelic drugs, using molecular dynamics simulations. Without Gqα, the active receptor collapses to an inactive conformation, underscoring the G‐protein's role. Discovery of an intermediate “partially‐open” state and the higher affinity of ligands to the orthosteric binding site provides ...

The role of host PrP in Transmissible Spongiform Encephalopathies [PDF]
PrP has a central role in the Transmissible Spongiform Encephalopathies (TSEs), and mutations and polymorphisms in host PrP can profoundly alter the host's susceptibility to a TSE agent.

Low‐density lipoprotein receptor‐related protein 6 (LRP6) is a key receptor for the Wnt antagonist Dickkopf1 (DKK1). DKK1 protein expression is induced in a bleomycin (BLM)‐induced lung injury model. We show that DKK1 induces proinflammatory and profibrotic genes in lung fibroblasts.
