Therapeutic strategies impacting cancer cell glutamine metabolism

open access: yes, 2013
The metabolic adaptations that support oncogenic growth can also render cancer cells dependent on certain nutrients. Along with the Warburg effect, increased utilization of glutamine is one of the metabolic hallmarks of the transformed state.

Optimized Ex Vivo Human Liver Slice Culture Maintains Extended Viability and Function for Hepatotoxicity Testing

open access: yesAdvanced Science, EarlyView.
An optimized ex vivo human liver slice culture system preserves tissue architecture, hepatocyte function, and immune‐stromal complexity for at least five days. By refining slice thickness, oxygenation, air‐liquid interface, and extracellular matrix support, the platform enables human‐relevant hepatotoxicity testing and mechanistic studies, offering a ...

Vascularized Liver‐on‐a‐Microsphere Reveals Alanine‐Glucose Metabolism‐Driven Regulation of Liver Function and Injury

open access: yesAdvanced Science, EarlyView.
A vascularized liver‐on‐a‐microsphere system is developed to recreate lobule‐like cellular organization and endothelial barrier function within compartmentalized hydrogel particles. The platform enhances hepatic performance and reveals alanine‐glucose cycle activation linked to improved detoxification capacity, offering a high‐throughput microscale ...

RBM12 Maintains Glioma Stem Cells by Activating Amino Acid‐Dependent mTORC1 Signaling via SLC7A5 mRNA Stabilization

open access: yesAdvanced Science, EarlyView.
This study shows that in glioma stem cells (GSCs), RBM12 recruits ALKBH5 to remove m6A from SLC7A5 transcripts, thereby enhancing mRNA stability, which elevates large neutral amino acid (LNAA) levels and activates mTORC1, promoting GSC proliferation, self‐renewal, and tumor growth.

Multi‐Component Functionalized Bifidobacterium Longum Hydrogel for Multi‐Target Integrated Therapy of Colitis‐Associated Anxiety and Depression

open access: yesAdvanced Science, EarlyView.
A pH/MMP dual‐responsive Bifidobacterium longum (BL) hydrogel (INPs@BL@Gel) functionalized with baicalin, tyrosine, and inulin is constructed. It enables gastric protection, inflammation‐targeted release, and prolonged intestinal colonization. By remodeling gut microbiota, elevating beneficial neuroactive metabolites (homovanillic acid, short‐chain ...

Light‐Activated Ruthenium Nanoclusters Reprogram the Metabolic‐Quorum Sensing Axis for Precision Periodontitis Therapy

open access: yesAdvanced Science, EarlyView.
Under 660 nm visible light, Ru NCs efficiently photocatalyze nitrate‐to‐ammonium conversion and modulate oral nitrogen metabolism. They precisely perturb Veillonella parvula, block its key support to Porphyromonas gingivalis, and exert an “inhibiting without killing” antivirulence effect.

Glutamine metabolism and its roles in tumor radiotherapy by regulating DNA damage repair

open access: yesFrontiers in Cell and Developmental Biology
BackgroundAs a conditionally essential amino acid that is required by tumor cells, glutamine serves as a key metabolic hub in tumor cells, functioning both as a carbon source for the tricarboxylic acid cycle and lipid synthesis and as a nitrogen source ...

REGγ Suppresses Ferroptosis and Induces Drug Resistance by Degrading WDR6 in Chondrosarcoma

open access: yesAdvanced Science, EarlyView.
Here, we identified REGγ as a susceptibility factor in chondrosarcoma. Our study demonstrates that abnormally activated REGγ‐20S proteasome promotes chondrosarcoma development and progression. Further validation in animal models revealed that blocking REGγ function induced ferroptosis, suppressed malignant progression of chondrosarcoma, and uncovered a

Targeting the crosstalk between glutamine metabolism and tumor immune microenvironment for lung cancer immunotherapy

open access: yesInterdisciplinary Medicine
Lung cancer remains one of the deadliest cancers globally, which may be attributed in part to a limited understanding of its development. Therefore, exploration of the fundamental processes of lung cancer and innovative therapies is imminent.
