Overview of glutamine metabolism in stromal components of the tumor microenvironment and potential anti-tumor therapies

open access: yesGenes and Diseases
As a critical metabolite in the tumor microenvironment, glutamine plays a crucial role in tumor progression, and its dual effects on promoting and inhibiting tumors have garnered increasing attention in recent years.

A glutamine metabolic switch supports erythropoiesis

open access: yesScience
Metabolic requirements vary during development, and our understanding of how metabolic activity influences cell specialization is incomplete. Here, we describe a switch from glutamine catabolism to synthesis required for erythroid cell maturation. Glutamine synthetase (GS), one of the oldest functioning genes in evolution, is activated during erythroid

ZER1 Restrains Pressure Overload‐Induced Cardiac Remodeling by Targeting DVL2 for Gly/N‐Degron‐Dependent Degradation

open access: yesAdvanced Science, EarlyView.
Pressure overload suppresses cardiomyocyte ZER1, weakening CRL2Zer1‐mediated DVL2 degradation and allowing DVL2 accumulation. Elevated DVL2 activates CaMKII‐HDAC4‐MEF2C signaling, drives fetal gene reactivation, and promotes pathological remodeling.

Metabolic adaptation of tumor cells under chronic acidosis: a shift towards reductive glutamine metabolism driven by the SIRT1/HIF2 axis

open access: yes, 2015
Cancer progression is strongly influenced by the physico-chemical properties of the tumor microenvironment. In particular, tumor cells must adapt to survive under low pO2 and low pH.

Integrative Omics Analysis Reveals the Regulation of Hypoxia Tolerance in Large Yellow Croaker (Larimichthys crocea) via the Lipoic Acid Synthase (lias) Gene

open access: yesAdvanced Science, EarlyView.
Lipoic acid synthase (lias) can regulate α‐KG levels through lipoylation, thereby negatively regulating HIF‐1α protein levels via PHD under hypoixa. The Hap2 allele of lias exhibits lower expression levels than Hap1, leading to the accumulation of more HIF‐1α protein and thereby enhancing hypoxia tolerance. ABSTRACT Hypoxia stress seriously affects the

Slc44a2 Deficiency Unveils an IFN‐I–Dependent Feedback Control of pDC Egress

open access: yesAdvanced Science, EarlyView.
Working model of SLC44A2‐mediated maintenance of pDC homeostasis. This model illustrates two central mechanisms by which SLC44A2 regulates pDC homeostasis: (1) SLC44A2 limits IFN‐I production by exporting amino acids (T, N, Q), thereby preventing spontaneous pDC activation.

Nanozyme‐Reinforced miR‐197‐3p Delivery Resets Metabolic and Senescence Pathways to Rejuvenate Osteoarthritic Cartilage

open access: yesAdvanced Science, EarlyView.
ABSTRACT Osteoarthritis (OA) is a progressive and disabling joint disease driven by oxidative stress, chondrocyte senescence and extracellular matrix (ECM) degradation, yet lacks effective disease‐modifying treatments. In this study, we identified miR‐197‐3p as a previously unrecognized, cartilage‐protective miRNA significantly downregulated in both ...
