Advances in research on the impact and mechanisms of pathogenic microorganism infections on pyroptosis

open access: yesFrontiers in Microbiology
Pyroptosis, also known as inflammatory necrosis, is a form of programmed cell death characterized by the activation of gasdermin proteins, leading to the formation of pores in the cell membrane, continuous cell swelling, and eventual membrane rupture ...

Targeted Cell-to-Cell Delivery of Protein Payloads via the Granzyme-Perforin Pathway

open access: yesMolecular Therapy: Methods & Clinical Development, 2017
There is great potential for engineering cellular therapeutics by repurposing biological systems. Here, we report utilization of the granzyme-perforin pathway of cytotoxic lymphocytes as a cell-to-cell protein delivery module. We designed and constructed

CD207‐Positive Dendritic Cells Promote Emphysema Through CD8+ T Cell Pathway in Chronic Obstructive Pulmonary Disease

open access: yesAdvanced Science, EarlyView.
CD207+ dendritic cells (DCs) drive emphysema by promoting CD8⁺ T cell cytotoxicity via Birbeck granule‐dependent MHC‐I antigen presentation. This DC subset is expanded by cigarette smoke‐induced oxidative stress, which triggers granulocyte‐macrophage colony‐stimulating factor (GM‐CSF) release from airway epithelium.

Tailoring CD19xCD3-DART exposure enhances T-cells to eradication of B-cell neoplasms. [PDF]

open access: yes, 2018
Many patients with B-cell malignancies can be successfully treated, although tumor eradication is rarely achieved. T-cell-directed killing of tumor cells using engineered T-cells or bispecific antibodies is a promising approach for the treatment of ...

Pro-inflammatory granzyme K contributes extracellularly to disease

open access: yesFrontiers in Immunology
Granzyme K (GzmK) is an immune-secreted serine protease typically expressed at low levels but elevated in response to tissue injury and disease. Known as an orphan granzyme due to limited scientific investigation, this tryptase is being redefined as ...

FAPα+ Macrophages Orchestrate Immune Evasion in Multiple Myeloma by Dual Regulation of PD‐L1 and T Cell Senescence

open access: yesAdvanced Science, EarlyView.
Fibroblast activation protein alpha‐positive (FAPα+) macrophages, a distinct subset of multiple myeloma (MM)‐associated macrophages, drive immune evasion in MM through multi‐faceted mechanisms. FAPα physically interacts with vimentin (VIM) and triggers its phosphorylation at the S72 residue, which in turn induces PD‐L1 transcription. Additionally, FAPα

Tailoring Vascular‐Immune Homeostasis via Manganese‐DNA Complex‐Armed Immunogenic Extracellular Vesicles for Pancreatic Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
This study demonstrates that Mn2⁺–tumor DNA complexes encapsulated in dendritic cell (DC)– derived immunogenic extracellular vesicles (EVDC@Mn‐DNA) act as a DC‐specific cGAS– STING activator. EVDC@Mn‐DNA treatment enhances intratumoral DC activation, improves tumor vascular function, promotes CD8⁺ T cell activity, and suppresses pancreatic tumor growth,

The role of tumor necrosis factor-α and natural killer cells in uterine artery function and pregnancy outcome in the stroke prone spontaneously hypertensive rat [PDF]

open access: yes, 2016
Women with chronic hypertension are at increased risk of maternal and fetal morbidity and mortality. We have previously characterized the stroke-prone spontaneously hypertensive rat (SHRSP) as a model of deficient uterine artery function and adverse ...

Tim-3 suppresses the killing effect of Vγ9Vδ2 T cells on colon cancer cells by reducing perforin and granzyme B expression.

open access: yesExperimental Cell Research, 2019
Gamma delta (γδ) T cell-based tumor immunotherapy has been one of the most promising cancer immunotherapeutic strategies. However, the key regulators of the Vγ9Vδ2 T cell-mediated antitumor response remain unclear.

PD‐1 Inhibits CD4+ TRM‐Mediated cDC1 Mobilization via Suppressing JAML in Human NSCLC

open access: yesAdvanced Science, EarlyView.
CD4+ tissue‐resident memory T cells (TRMs) in non‐small cell lung cancer recruit conventional type 1 dendritic cells via XCL1‐XCR1 signaling, orchestrating antitumor immunity. The costimulatory molecule JAML is essential for this process. PD‐1 blockade restores JAML expression and cDC1 mobilization, while JAML agonists synergize with anti‐PD‐1 therapy,
