Chemical-induced dimerization of GSDMD C-terminal domain blocks GSDMD N-terminal domain-mediated pyroptosis

Abstract GSDMD is a pyroptosis executioner protein, in which the C-terminal domain (GD-CT) inhibits the pore-forming ability of N-terminal domain (GD-NT). Caspase-cleavage separates GD-NT and GD-CT, and then the free GD-NTs oligomerize and form nano-sized pyroptotic pores in plasma membrane.

PD alleviates APAP‐induced ALI by inhibiting the NLRP3 inflammasome signaling pathway. C/EBPβ is identified as a direct target via multimethod validation. PD promotes the deacetylation of C/EBPβ via Sirt1. The Sirt1/C/EBPβ/NLRP3 signaling axis is defined for PD's hepatoprotective effect.

S100A9 promotes pulmonary arterial hypertension by regulating mitochondria–endoplasmic reticulum interaction‐mediated inflammatory injury of endothelial cells

Macrophage‐derived S100 calcium‐binding protein A9 (S100A9) promotes the pathological progression of pulmonary arterial hypertension (PAH). S100A9 upregulates the interaction between signal‐transducing adaptor protein 2 and leucine‐rich repeat kinase 2, thereby regulating mitochondria–endoplasmic reticulum (ER) contact.
