Chemical Biology &Drug Design, Volume 107, Issue 4, April 2026.Electrophilic warheads were incorporated into Hsp90β‐selective inhibitors to covalently modify Lys58, which occupies the solvent‐exposed region of the binding pocket. Although many inhibitors display nanomolar affinity towards Hsp90β over a 24‐h period, analyses to determine covalent bond formation with this residue were inconclusive.
