Reframing Alzheimer's disease as a complex adaptive system: More than an amyloid beta–tau connection
Abstract Alzheimer's disease (AD) is the leading cause of dementia, but simple models focusing on amyloid beta and tau only partially explain its variability and limited success in treatment. Evidence from systems biology, neuroimmunology, connectomics, and computational modeling supports viewing AD as a complex adaptive system, a multiscale network in

Attempts to Achieve Targeted Covalent Inhibition of Hsp90β
Electrophilic warheads were incorporated into Hsp90β‐selective inhibitors to covalently modify Lys58, which occupies the solvent‐exposed region of the binding pocket. Although many inhibitors display nanomolar affinity towards Hsp90β over a 24‐h period, analyses to determine covalent bond formation with this residue were inconclusive.

The NLRP3 Inflammasome: Mechanisms of Activation, Regulation, and Therapeutic Opportunities
Diverse diseases converge on NLRP3. We depict a discovery‐to‐clinic track: high‐throughput/phenotypic screens, structure‐guided design, and modality innovation (allosteric inhibitors, interface blockers, degraders) deliver third‐generation, disease‐tailored NLRP3 control.

BioE is a new diiron oxygenase that catalyzes the conversion of long‐chain acyl groups into pimeloyl thioester, initiating biotin synthesis. The overexpression of EmBioE disrupts lipid metabolic homeostasis, requiring repressor BioL to maintain a balance between long‐chain fatty acids and biotin synthesis.
