A Depolarizing Leak in Sodium Bicarbonate Cotransporter NBCe1 Causes Brain Edema

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objectives SLC4A4 encodes electrogenic sodium bicarbonate cotransporter NBCe1, prominently expressed in kidney and brain. Recessive loss‐of‐function variants in SLC4A4 cause proximal renal tubular acidosis, no brain edema. In the brain, NBCe1 is expressed by astrocytes, where it regulates pH and mediates astrocyte volume changes.

Overexpression of G-protein-coupled receptor 40 enhances the mitogenic response to epoxyeicosatrienoic acids. [PDF]

open access: yesPLoS ONE, 2015
The cytochrome P450 epoxygenase-dependent arachidonic acid metabolites, epoxyeicosatrienoic acids (EETs), are potent survival factors and mitogens for renal epithelial cells, but the molecular identity in the cells that initiates the mitogenic signaling ...

Scalable Production of AAV Vectors in Orbitally Shaken HEK293 Cells [PDF]

open access: yesMolecular Therapy - Methods & Clinical Development, 2019
Adeno-associated virus (AAV) vectors are currently among the most commonly applied for in vivo gene therapy approaches. The evaluation of vectors during clinical development requires the production of considerable amounts of highly pure and potent vectors.

SOAT expression in stably transfected SOAT-HEK293 cells.

open access: yes, 2013
In the SOAT-HEK293 cells SOAT expression was induced by pre-treatment with tetracycline (+Tet). Control cells were untreated with tetracycline (−Tet) or represent non-transfected HEK293 cells (contr.). (A) Cell lysates were processed for WB analysis with

Ofatumumab in Myelin Oligodendrocyte Glycoprotein Antibody–Associated Disease: A Comparison With Rituximab

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective To evaluate the efficacy and safety of ofatumumab in patients with myelin oligodendrocyte glycoprotein antibody–associated disease (MOGAD), and compare it with rituximab. Methods We conducted a single–center, observational study including 22 MOGAD patients treated with ofatumumab and 21 treated with rituximab.

ATP homeostasis is maintained by AMPK-FOXO-CKB pathway in HEK293 cells under oxidative stress conditions.

open access: yes, 2023
(a) Knockdown of CKB or AMPKα1/α2 mRNA expression did not affect the basal ATP content in HEK293 cells. (b) Knockdown of CKB or AMPKα1/α2 mRNA expression increased the fold of ATP decrease caused by paraquat treatment for 24h in HEK293 cells.

Comparative Effectiveness and Safety of Inebilizumab Versus Rituximab in AQP4‐IgG‐Positive NMOSD

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Rituximab (anti‐CD20, RTX) and inebilizumab (anti‐CD19, INE) represent B‐cell‐depleting therapies used for aquaporin‐4 antibody‐positive (AQP4‐IgG+) neuromyelitis optica spectrum disorder (NMOSD); however, direct comparative evidence remains limited.

Knocking out Ornithine Decarboxylase Antizyme 1 (OAZ1) Improves Recombinant Protein Expression in the HEK293 Cell Line

open access: yesMedical Sciences, 2018
Creating efficient cell lines is a priority for the biopharmaceutical industry, which produces biologicals for various uses. A recent approach to achieving this goal is the use of non-coding RNAs, microRNA (miRNA) and small interfering RNA (siRNA), to ...

Methyl-Cantharidimide (MCA) Has Anticancer Efficacy in ABCB1- and ABCG2-Overexpressing and Cisplatin Resistant Cancer Cells

open access: yesFrontiers in Oncology, 2020
In this study, we investigated the efficacy of methyl-cantharidimide (MCA), a cantharidin (CTD) analog, as an anticancer drug, in cancer cells overexpressing either ABCB1 or ABCG2 transporters and in cisplatin-resistant cancer cells.

HSP70 stabilizes TRIM63-containing CBs in HEK293 cells.

open access: yes, 2017
(A) HEK293 cells were transiently transfected with FLAG-TRIM63 (+) or vector alone (-). The association of TRIM63 with endogenous HSP70 was analyzed with co-immunoprecipitation followed by western blotting (WB).
