The amino acid sequence of human hexokinase II was deduced from the sequence of cDNA clones isolated from a skeletal muscle library. An open reading frame of 2751 bases encodes a protein of 917 amino acids. The deduced amino acid sequence has 94% identity with rat hexokinase II but only 72% identity with human hexokinase type I.

Abstract A batch chromatographic method for the determination of hexokinase employing trace-labelled glucose and resin chromatography is described. Markedly enhanced sensitivity compared with the standard spectrophotometric assay is shown. Its greater reliability and use for particulate-tissue preparations is discussed.

The anomeric specificity of hexokinase was examined in crude homogenates of rat parotid gland, erythrocytes and pancreatic islets. At 8 degrees C, the alpha/beta ratio in maximal velocity averaged 0.73, 0.66 and 0.75 in the parotid, erythrocytes and pancreatic islets, respectively.

Abstract Clear crystalline lenses were obtained post-mortem from human prenates, neonates, young and old adults. Senile cataracts removed surgically were also studied. The specific activity of soluble fetal lens hexokinase is equal to that of the rat lens. There is a progressive decrease in specific activity as the lens size increases.
