The highly conserved eukaryotic histone chaperone ASF1 is involved in the assembly and disassembly of nucleosomes during transcription, DNA replication, and repair. It was the first chaperone discovered to be involved in all three of these processes. The

Cell-fate maintenance is important to preserve the variety of cell types that are essential for the formation and function of tissues. We previously showed that the acetylated histone-binding protein BET-1 maintains cell fate by recruiting the histone ...

Hydroxamic Acids as HDAC Inhibitor Drug Leads for Malaria
ABSTRACT Malaria is a global health threat, with an estimated 282 million cases and 610,000 malaria‐associated deaths reported in 2024. Most mortality is due to infection by Plasmodium falciparum parasites, with the highest burden occurring in Sub‐Saharan Africa.

Regulation of histone acetylation and turnover by histone chaperones
Acetylation of histone H3 on lysine 56 occurs on newly synthesized histones in fungal species, and the absence of this modification is associated with extreme sensitivity to genotoxic agents. H3‐K56 acetylation is catalyzed when Rtt109, a distant homolog of p300/CBP acetyltransferases, is stimulated by either ...

Role of Template Activating Factor-I as a chaperone in linker histone dynamics
Linker histone H1 is a fundamental chromosomal protein involved in the maintenance of higher-ordered chromatin organization. The exchange dynamics of histone H1 correlates well with chromatin plasticity.

Abstract Myotonic dystrophy type 1 (DM1) is a clinically challenging multisystem neuromuscular hereditary disorder, with generational increase in severity and earlier age at onset. It is caused by an unstable cytosine‐thymine‐guanine repeat expansion at the DMPK locus, accompanied by associated genetic and epigenetic modifications.

Histone chaperone Nap1 facilitates histone dynamics in the nucleosome
We developed a single‐molecule FRET (smFRET) experimental system with which we can separately monitor histone H2A‐H2B dimer dynamics and DNA dynamics within a nucleosome particle. Utilizing the system, we monitored the nucleosome dynamics in the presence of histone chaperone Nap1 in order to investigate how the chaperone alters the dynamics of the ...

Multiple Roles of Protamine Kinase SRPK1 and Phosphatase PP1γ in Sperm Development
ABSTRACT Protein phosphorylation regulates key events of male germ cell differentiation. In the testis, SRPK1 and PP1γ are involved in protamine phosphorylation and dephosphorylation, respectively, and their deregulation is linked to defective chromatin structure and male infertility.

The Role of the MCM2-7 Helicase Subunit MCM2 in Epigenetic Inheritance
Recycling histone proteins from parental chromatin, a process known as parental histone transfer, is an important component in chromosome replication and is essential for epigenetic inheritance.

Cytoplasmic DAXX drives SQSTM1/p62 phase condensation to activate Nrf2-mediated stress response
The autophagy protein p62 undergoes liquid-liquid phase separation but how this is regulated is unclear. Here, the authors report that the histone chaperone DAXX interacts with p62 in the cytoplasm to drive its phase separation.
