Activation of AMP-activated protein kinase by metformin induces protein acetylation in prostate and ovarian cancer cells [PDF]
AMP-activated protein kinase (AMPK) is an energy sensor and master regulator of metabolism. AMPK functions as a fuel gauge monitoring systemic and cellular energy status.

Sites of Histone/Histone Interaction in the H3 · H4 Complex [PDF]
Sites of interaction between histones H3 and H4 have been probed by investigating complex formation, firstly between histone H4 and three peptides cleaved by chemical means from histone H3 (residues 1–90 and 1–120 using cyanogen bromide and residues 42–135 using N‐bromosuccinimide), secondly between histone H3 and two peptides cleaved from histone H4 ...

Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway
Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.

Histone H3 tail clipping regulates gene expression [PDF]
Induction of gene expression in yeast and human cells involves changes in the histone modifications associated with promoters. Here we identify a histone H3 endopeptidase activity in Saccharomyces cerevisiae that may regulate these events. The endopeptidase cleaves H3 after Ala21, generating a histone that lacks the first 21 residues and shows a ...

HDAC4 is degraded by the E3 ligase FBXW7. In colorectal cancer, FBXW7 mutations prevent HDAC4 degradation, leading to oxaliplatin resistance. Forced degradation of HDAC4 using a PROTAC compound restores drug sensitivity by resetting the super‐enhancer landscape, reprogramming the epigenetic state of FBXW7‐mutated cells to resemble oxaliplatin ...

Histone modification changes upon exposure of human lung adenocarcinoma cells to nanoparticles
The increasing use of nanoparticles in numerous applications has led to growing concern about their potential toxicological properties. However, the role of epigenetic alterations in toxicity due to nanoparticles remains relatively unexplored.

Chromatin status and transcription factor binding to gonadotropin promoters in gonadotrope cell lines. [PDF]
BackgroundProper expression of key reproductive hormones from gonadotrope cells of the pituitary is required for pubertal onset and reproduction. To further our understanding of the molecular events taking place during embryonic development, leading to ...

Structural cooperativity in histone H3 tail modifications [PDF]
AbstractPost‐translational modifications of histone H3 tails have crucial roles in regulation of cellular processes. There is cross‐regulation between the modifications of K4, K9, and K14 residues. The modifications on these residues drastically promote or inhibit each other.

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...
