The human neurotropic polyomavirus, JC virus (JCV), is the etiologic agent of progressive multifocal leukoencephalopathy (PML), a fatal demyelinating disease of the central nervous system that occurs mainly in immunosuppressed patients. JCV has also been found to be associated with human tumors of the brain and other organs.

John Cunningham virus (JCV) is a double-stranded DNA polyomavirus isolated in 1971 by Padgett et al. from the brain of a patient with Hodgkin disease. Viral DNA is transcripted on both strands and it encodes for the early genes counterclockwise and for the late genes clockwise.
