A monoclinic polymorph of hypoxanthine (systematic name: 1,7-dihydro-6H-purin-6-one), C5H4N4O, is reported. The hydrogen-bonding motifs involve a layered structure of N—H⋯O and N—H⋯N hydrogen-bonded mol­ecules. The mean stacking distance between adjacent layers is 3.672 A.
