Illumina MiSeq sequencing disfavours a sequence motif in the GFP reporter gene [PDF]

open access: yesScientific Reports, 2016
AbstractGreen fluorescent protein (GFP) is one of the most used reporter genes. We have used next-generation sequencing (NGS) to analyse the genetic diversity of a recombinant influenza A virus that expresses GFP and found a remarkable coverage dip in the GFP coding sequence.

A Short Review on Illumina Sequencing Technology

open access: yesAustin Journal of Veterinary Science & Animal Husbandry, 2022
DNA sequencing is that the method of crucial the supermolecule sequence – the order of nucleotides in deoxyribonucleic acid. It includes any methodology or technology that’s accustomed verify the order of the four bases: A, guanine, cytosine, and T. Illumina technology is sequencing by synthesis approach and is presently the foremost used technology ...

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

A Comparison of Base-calling Algorithms for Illumina Sequencing Technology [PDF]

open access: yesBriefings in Bioinformatics, 2015
Recent advances in next-generation sequencing technology have yielded increasing cost-effectiveness and higher throughput produced per run, in turn, greatly influencing the analysis of DNA sequences. Among the various sequencing technologies, Illumina is by far the most widely used platform.

Diagnostic applications of next generation sequencing: working towards quality standards [PDF]

open access: yes, 2012
Over the past 6 years, next generation sequencing (NGS) has been established as a valuable high-throughput method for research in molecular genetics and has successfully been employed in the identification of rare and common genetic variations. All major

MITF maintains genome stability in nonmelanocyte lineages

open access: yesMolecular Oncology, EarlyView.
MITF is essential for melanocyte survival and acts as an oncogene in 10%–20% of melanomas. We show that MITF depletion causes genome instability in nonmelanocytic cells, leading to LATS2‐mediated P53 activation, cell cycle arrest, and apoptosis. This study highlights the role of MITF as a genome maintenance factor beyond the melanocyte lineage. Created

A large genome center's improvements to the Illumina sequencing system [PDF]

open access: yesNature Methods, 2008
The Wellcome Trust Sanger Institute is one of the world's largest genome centers, and a substantial amount of our sequencing is performed with 'next-generation' massively parallel sequencing technologies: in June 2008 the quantity of purity-filtered sequence data generated by our Genome Analyzer (Illumina) platforms reached 1 terabase, and our average ...
