This article describes a regulatory circuit in Escherichia coli able to detect protein–protein interactions with exquisite sensitivity. The interaction between two hybrid proteins fused to Bordetella pertussis adenylate cyclase and its activator calmodulin triggers a potent cyclic AMP signaling cascade.

A genome‐wide microRNA CRISPR screen identifies miR‐18a as a master regulator of cross‐resistance in melanoma. Loss of miR‐18a activates the AJUBA–YAP/Hippo axis to confer BRAFi resistance and enhances THBS1–CD47 interaction to impair CD8+ T cell immunity. hnRNP A1 is identified as an upstream regulator of miR‐18a processing.

Causal Prediction of TP53 Variant Pathogenicity Using a Perturbation‐Informed Protein Language Model
A TP53‐specific predictor, CaVepP53, is developed by fine‐tuning ESMC on experimentally validated variants, quantifying pathogenicity via Euclidean distances. It outperforms general‐purpose models and extends to five cancer genes, enabling interpretable variant classification for precision medicine.
