An in Vitro Microenvironment for Nucleus Pulposus Regeneration

open access: yes, 2019
Intervertebral disc degeneration begins early in life, increases with age, and can ultimately result in disc failure. At the center of the disc, the nucleus pulposus (NP) resides in a hypoxic environment where degeneration is thought to begin. Initially, nucleus pulposus cells (NPCs) display altered morphology and function, along with increased rates ...

Tumor‐stromal crosstalk and macrophage enrichment are associated with chemotherapy response in bladder cancer

open access: yesFEBS Open Bio, EarlyView.
Chemoresistance in bladder cancer: Macrophage recruitment associated with CXCL1, CXCL5 and CXCL8 expression is characteristic of Gemcitabine/Cisplatin (Gem/Cis) Non‐Responder tumors (right side) while Responder tumors did not show substantial tumor‐stromal crosstalk (left side). All biological icons are attributed to Bioicons: carcinoma, cancerous‐cell‐

Scaffolds Mimicking the Tumor Microenvironment for In Vitro Malignancy Models

open access: yesBiomimetics
The tumor microenvironment (TME) plays a crucial role in regulating cancer cell proliferation, invasion, and drug resistance. Traditional two-dimensional (2D) in vitro models and animal models often fail to replicate the biochemical and biophysical ...

Microglial dynamics and ferroptosis induction in human iPSC‐derived neuron–astrocyte–microglia tri‐cultures

open access: yesFEBS Open Bio, EarlyView.
A tri‐culture of iPSC‐derived neurons, astrocytes, and microglia treated with ferroptosis inducers as an Induced ferroptosis model was characterized by scRNA‐seq, cell survival, and cytokine release assays. This analysis revealed diverse microglial transcriptomic changes, indicating that the system captures key aspects of the complex cellular ...

In vitro 3D modeling of colorectal cancer: the pivotal role of the extracellular matrix, stroma and immune modulation

open access: yesFrontiers in Genetics
Colorectal cancer (CRC) is a leading global cancer with high mortality, especially in metastatic cases, with limited therapeutic options. The tumor microenvironment (TME), a network comprising various immune cells, stromal cells and extracellular (ECM ...

Identification of functional murine mitochondrial formyl peptides and their effects on myeloid‐derived suppressor cell generation

open access: yesFEBS Open Bio, EarlyView.
We first identified functional murine mitochondrial N‐formyl peptides (MT‐FPs) and investigated their effects on the in vitro myeloid‐derived suppressor cell (MDSC) generation from bone marrow cells. We demonstrated that MT‐FPs acted directly on bone marrow cells to promote MDSC generation and modulated the polymorphonuclear (PMN)‐MDSC/monocyte (M ...

Exon 7 splicing of ERα predicts poor prognosis and increases phenotypic heterogeneity in luminal a subtype breast cancer

open access: yesFEBS Open Bio, EarlyView.
ERα splice variant ERα∆7 lacks the C‐terminus, and its expression may change phenotypes of breast cancers. Our results showed that ERα∆7 is found in the luminal A subtype, and elevated ERα∆7 levels are linked to improved cell survival with lower proliferation and migration.

A novel organoid model retaining the glioma microenvironment for personalized drug screening and therapeutic evaluation

open access: yesBioactive Materials
Glioma is an aggressive brain tumor with a poor prognosis. Establishing an in vitro culture model that closely replicates the cellular composition and microenvironment of the original tumor has been challenging, limiting its clinical applications.
