A Quantum Model on Chemically-Physically Induced Pluripotency in Stem Cells [PDF]

open access: yesarXiv, 2013
A quantum model on the chemically and physically induced pluripotency in stem cells is proposed. Based on the conformational Hamiltonian and the idea of slow variables (molecular torsions) slaving fast ones the conversion from the differentiate state to pluripotent state is defined as the quantum transition between conformational states.

Transcriptional profiling reveals fundamental differences in iPS-derived progenitors of endothelial cells (PECs) versus adult circulating EPCs [PDF]

open access: yesarXiv, 2020
There are a number of different stem cell sources that have the potential to be used as therapeutics in vascular degenerative diseases. On the one hand, there are so called endothelial progenitor cells (EPCs), which are typically derived from adult blood. They carry the marker CD34, but the true nature and definition of EPCs is still controversial.

A Visible Light‐Responsive Hydrogel to Study the Effect of Dynamic Tissue Stiffness on Cellular Mechanosensing

open access: yesAdvanced Functional Materials, EarlyView.
A visible light‐responsive polyacrylamide‐azobenzene hydrogel enables safe, reversible stiffness control for studying cell mechanobiology without harmful UV exposure. This approach reveals stem cells respond rapidly to mechanical changes, showing altered shape and protein distribution within one hour.

Theoretical modelling discriminates the stochastic and deterministic hypothesis of cell reprogramming [PDF]

open access: yesarXiv, 2014
How to induce differentiated cells into pluripotent cells has elicited researchers' interests for a long time since pluripotent stem cells are able to offer remarkable potential in numerous subfields of biological research. However, the nature of cell reprogramming, especially the mechanisms still remain elusive for the sake of most protocols of ...

Machine Learning Guided Design of Nerve‐On‐A‐Chip Platforms with Promoted Neurite Outgrowth

open access: yesAdvanced Functional Materials, EarlyView.
Compared to labor‐intensive trial‐and‐error experimentation, a machine learning (ML)‐guided workflow, incorporating cell viability assays, data augmentation, ensemble modeling, and model interpretation, is developed to accelerate nerve‐on‐a‐chip optimization and uncover data‐driven design principles.

Dynamic full-field optical coherence tomography: 3D live-imaging of retinal organoids [PDF]

open access: yes, 2019
Optical coherence tomography offers astounding opportunities to image the complex structure of living tissue, but lacks functional information. We present dynamic full-field optical coherence tomography to image living human induced pluripotent stem cell-derived retinal organoids non-invasively.

Pericyte‐Assisted Vascular Lumen Organization in a Novel Dynamic Human Blood‐Brain Barrier‐on‐Chip Model

open access: yesAdvanced Healthcare Materials, EarlyView.
Organ‐on‐Chip (OoC) technology enables the precise modeling of the blood‐brain barrier (BBB). This study presents a microfluidic device that mimics brain microvessels, integrating co‐culture with pericytes and astrocytes under pulsatile flow. Results show that perivascular cells modulate endothelial morphology, promoting lumen formation and barrier ...

The recent advances in the mathematical modelling of human pluripotent stem cells [PDF]

open access: yesarXiv, 2019
Human pluripotent stem cells hold great promise for developments in regenerative medicine and drug design. The mathematical modelling of stem cells and their properties is necessary to understand and quantify key behaviours and develop non-invasive prognostic modelling tools to assist in the optimisation of laboratory experiments.

Parallel derivation of isogenic human primed and naive induced pluripotent stem cells

open access: yesNature Communications, 2018
Induced pluripotent stem cells (iPSCs) have considerably impacted human developmental biology and regenerative medicine, notably because they circumvent the use of cells of embryonic origin and offer the potential to generate patient-specific pluripotent

Soft Pneumatic Device Designed to Mimic the Periosteal Environment for Regulating the Fate of Mesenchymal Stem Cells

open access: yesAdvanced Healthcare Materials, EarlyView.
This study presents a custom‐made soft pneumatic device (SPD) that mimics complex mechanical forces and fluid flow to enhance in vitro cell differentiation. It shows that low mechanotransduction promotes MSC differentiation into osteoprogenitor cells, while high mechanotransduction causes structural disruptions and cell detachment, providing new ...
