A simple method for developing an infectious cDNA clone of Japanese encephalitis virus

Virus Genes, 2016
Flavivirus cDNA clones frequently demonstrate genetic instability in transformed bacteria, which hampers the construction and manipulation of cDNAs for infectious flaviviruses. In this study, we developed a stable, full-length cDNA clone, pJEHEN, of a GI JEV strain HEN0701 using a medium-copy-number pBR322 vector and propagating cDNA clones at room ...

Infectious in Vitro transcripts from a plum pox potyvirus cDNA clone

Virology, 1990
A full-length cDNA clone of the 9786 nt plum pox virus (PPV) RNA genome has been cloned downstream from a phage T7 RNA polymerase promoter. The RNAs synthesized by in vitro run-off transcription in the presence of the 5' cap analog m7GpppG were infectious in Nicotiana clevelandii plants.

In vitro mutagenesis of infectious viroid cDNA clone.

Journal of biochemistry, 1986
Previously we constructed an infectious hop stunt viroid (HSV) cDNA clone, PHS-P2P, which carries two copies of full-length HSV cDNA tandemly and generates HSV RNA when it is inoculated into cucumber plants. The in vitro transcript of the cDNA clone was also infectious.

Development and Characterization of an Infectious cDNA Clone of Equine Arteritis Virus

2017
Development and characterization of several infectious cDNA clones of equine arteritis virus (EAV) have been described in the literature. Here we describe the assembly of the full-length infectious cDNA clone of the virulent Bucyrus strain (VBS; ATCC VR-796) of EAV in a plasmid vector.

Characterization of infectious transcripts from a potato virus X cDNA clone

Virology, 1990
A full-length cDNA clone of potato virus X (PVX) has been constructed and fused to the bacteriophage T7 promoter in an in vitro transcription vector. Transcripts derived from this template (pMON 8660) were infectious when inoculated onto the local lesion host, Chenopodium amaranticolor.

In Vitro Transcription of Infectious Viral RNA from Cloned cDNA

1986
Publisher Summary This chapter describes the use of pPM1 as a vector for viral complementary DNA (cDNA) cloning and for in vitro production of infectious transcripts. The use of recombinant DNA technology in the study of most RNA viruses is limited by inability to express infection from viral cDNA clones.

Symptomless infectious cDNA clone of a Norwegian isolate of Poinsettia mosaic virus

Archives of Virology, 2008
An infectious cDNA clone of a Norwegian isolate of Poinsettia mosaic virus (PnMV) was generated. It consisted of 6,098 nucleotides and encoded a polyprotein of 219.5 kDa. Sequence comparisons indicated that this isolate shared 98.6% (nucleotide) and 97.1% (amino acid) identity with the previously sequenced isolate from Germany.

Construction and verification of an infectious cDNA clone of encephalomyocarditis virus from pigs

Journal of Virological Methods
In this study, a novel Encephalomyocarditis virus (EMCV) reverse genetic operating system was developed utilizing CMV promoters, enabling EMCV genome expression under the transcriptional control of the CMV immediate early promoter and BGH polyA transcriptional-termination signal.

Generation of Infectious Poliovirus with Altered Genetic Information from Cloned cDNA

2016
The effect of specific genetic alterations on virus biology and phenotype can be studied by a great number of available assays. The following method describes the basic protocol to generate infectious poliovirus with altered genetic information from cloned cDNA in cultured cells. The example explained here involves generation of a recombinant poliovirus
